The underlying pathophysiology of liver dysfunction in urea cycle disorders(UCDs) is still largely elusive. There is some evidence that the accumulation of urea cycle(UC) intermediates are toxic for hepatocyte mitochondria. It is possible that liver injury is directly caused by the toxicity of ammonia. The rarity of UCDs, the lack of checking of iron level in these patients, superficial knowledge of UC and an underestimation of the metabolic role of fumaric acid, are the main reasons that are responsible for the incomprehension of the mechanism of liver injury in patients suffering from UCDs. Owing to our routine clinical practice to screen for iron overload in severely ill neonates, with the focus on the newborns suffering from acute liver failure, we report a case of citrullinemia with neonatal liver failure and high blood parameters of iron overload. We hypothesize that the key is in the decreased-deficient fumaric acid production in the course of UC in UCDs that causes several sequentially intertwined metabolic disturbances with final result of liver iron overload. The presented hypothesis could be easily tested by examining the patients suffering from UCDs, for liver iron overload. This could be easily performed in countries with a high population and comprehensive national register for inborn errors of metabolism. Conclusion: Providing the hypothesis is correct, neonatal liver damage in patients having UCD can be prevented by the supplementation of pregnant women with fumaric or succinic acid, prepared in the form of iron supplementation pills. After birth, liverdamage in patients having UCDs can be prevented by supplementation of these patients with zinc fumarate or zinc succinylate, as well. 展开更多

Glucose metabolic phenotype of pancreatic cancer 预览

AIM: To construct a global 'metabolic phenotype' of pancreatic ductal adenocarcinoma(PDAC) reflecting tumour-related metabolic enzyme expression.METHODS: A systematic review of the literature was performed using Ovid SP and Pub Med databases using keywords 'pancreatic cancer' and individual glycolytic and mitochondrial oxidative phosphorylation(MOP) enzymes. Both human and animal studies investigating the oncological effect of enzyme expression changes and inhibitors in both an in vitro and in vivo setting were included in the review. Data reporting changes in enzyme expression and the effects on PDAC cells, such as survival and metastatic potential, were extracted to construct a metabolic phenotype. RESULTS: Seven hundred and ten papers were initially retrieved, and were screened to meet the review inclusion criteria. 107 unique articles were identified as reporting data involving glycolytic enzymes, and 28 articles involving MOP enzymes in PDAC. Data extraction followed a pre-defined protocol. There is consistent over-expression of glycolytic enzymes and lactate dehydrogenase in keeping with the Warburg effect to facilitate rapid adenosine-triphosphate production from glycolysis. Certain isoforms of these enzymes were over-expressed specifically in PDAC. Altering expression levels of HK, PGI, FBA, enolase, PK-M2 and LDA-A with metabolic inhibitors have shown a favourable effect on PDAC, thus identifying these as potential therapeutic targets. However, the Warburg effect on MOP enzymes is less clear, with different expression levels at different points in the Krebs cycle resulting in a fundamental change of metabolite levels, suggesting that other essential anabolic pathways are being stimulated. CONCLUSION: Further characterisation of the PDAC metabolic phenotype is necessary as currently there are few clinical studies and no successful clinical trials targeting metabolic enzymes. 展开更多

目的:探讨星形胶质细胞内的三羧酸循环在福尔马林诱导的大鼠炎性持续性痛、慢性痛和脊髓中枢敏化中的作用。方法:在大鼠右后肢足底注射福尔马林(5%,0.05 ml)制备炎性持续性痛大鼠模型,鞘内注射100 nmol/ml氟代柠檬酸(fluorocitrate... 目的:探讨星形胶质细胞内的三羧酸循环在福尔马林诱导的大鼠炎性持续性痛、慢性痛和脊髓中枢敏化中的作用。方法:在大鼠右后肢足底注射福尔马林(5%,0.05 ml)制备炎性持续性痛大鼠模型,鞘内注射100 nmol/ml氟代柠檬酸(fluorocitrate,FC)和/或5×10^4nmol/ml谷氨酸(L-glutamate,Glu)后,观察大鼠的行为学变化。结果:(1)急性期:与对照组相比,鞘内注射FC对大鼠自发伤害性行为(舔咬爪和缩腿反射)有抑制作用,而鞘内注射了Glu部分翻转了该抑制效应;(2)在慢性期,与对照组相比较,单次鞘内注射FC在3 h~2 d的时间点上显著提高大鼠同侧的50%爪缩阈值(P〈0.01,P〈0.05),而对侧50%爪缩阈值仅在第1 d时间点显示提高(P〈0.05)。随后在福尔马林注射后第9 d,再次鞘内注射FC,与对照组相比,能在3 h提高大鼠的同侧和对侧的50%爪缩阈值(P〈0.05),而在6 h阈值恢复到对照组水平(P〉0.05)。多次鞘内注射FC后,能够在3~7 d时间点上显著提高大鼠同侧的50%爪缩阈值(P〈0.01,P〈0.05),在2~7 d时间点上显著提高大鼠对侧的50%爪缩阈值(P〈0.01,P〈0.05)。随后在福尔马林注射后第9、10、11 d连续3 d鞘内注射FC,大鼠同侧的50%爪缩阈值的提高仅在鞘内注射日当天发生(P〈0.01,P〈0.05),次日即恢复到对照组水平(P〉0.05),而对侧的50%爪缩阈值在鞘内注射日第11 d及第12 d有所提高(P〈0.05),第13 d恢复到对照组水平(P〉0.05)。结论:星形胶质细胞内的三羧酸循环参与福尔马林诱导的急性痛和慢性痛的形成,但是在慢性痛的维持方面不起主导作用。 展开更多

Effect of Nitric Oxide on the Interaction Between Mitochondrial Malate Dehydrogenase and Citrate Synthase 预览

Mitochondrial malate dehydrogenase(m MDH) and citrate synthase(CS) are sequential enzymes in Krebs cycle. m MDH, CS and the complex between m MDH and CS(m MDH+CS) were treated with nitric oxide solution. The roles of notric oxide(NO) on the secondary structures and the interactions between m MDH and CS were studied using circular diehroism(CD) and Fourier transform surface plasmon resonance(FT-SPR), respectivley. The effects of NO on the activities of mM DH, CS and mM DH+CS were also studied. And the regulations by NO on m MDH and CS were simulated by Py MOL software. The results of SPR confirmed that strong interaction between m MDH and CS existed and NO could significantly regulate the interaction between the two enzymes. NO reduced the mass percents of α-helix and increased that of random in m MDH, CS and m MDH+CS. NO increased the activities of CS and m MDH+CS, and inhibited the activity of m MDH. Graphic simulation indicated that covalent bond was formed between NO and Asn242 in active site of CS. However, there was no direct bond between NO and m MDH. The increase in activity of mM DH+CS complex depended mostly on the interaction between NO and CS. All the results suggested that the regulations by NO on the activity and interaction between m MDH and CS were accord with the changes in m MDH, CS and m MDH+CS caused by NO. 展开更多
