Regularization of the level-set(LS)field is a critical part of LS-based topology optimization(TO)approaches.Traditionally this is achieved by advancing the LS field through the solution of a Hamilton-Jacobi equation combined with a reinitialization scheme.This approach,however,may limit the maximum step size and introduces discontinuities in the design process.Alternatively,energy functionals and intermediate LS value penalizations have been proposed.This paper introduces a novel LS regularization approach based on a signed distance field(SDF)which is applicable to explicit LSbased TO.The SDF is obtained using the heat method(HM)and is reconstructed for every design in the optimization process.The governing equations of the HM,as well as the ones describing the physical response of the system of interest,are discretized by the extended finite element method(XFEM).Numerical examples for pro?blems modeled by linear elasticity,nonlinear hyperelasticity and the incompressible Navier-Stokes equations in two and three dimensions are presented to show the applicability of the proposed scheme to a broad range of design optimization problems. 展开更多

Metastasis is the main cause of cancer death,and tumor cells mainly disseminate to the distal organs through the blood circulation,in which they experience considerable levels of fluid shear stress.CTCs are heterogeneous with diverse subpopulations of distinct genotypes and phenotypes and the frequency of CTCs is correlated with poor prognosis and overall survival in cancer patients.Less than 0.01%of them may eventually generate metastatic tumors in secondary sites,indicating the inefficiency of metastasis.Nevertheless,metastasis accounts for over 90%of cancer-related deaths,suggesting that a subpopulation of CTCs are able to survive the metastatic process and form metastases.To target metastasis,it is thus essential to understand the roles of various factors during dissemination in the survival and functions of CTCs.However,the effects of hemodynamic shear stress on biophysical properties and functions of CTCs in suspension are not fully understood.This study was to investigate the effect of hemodynamic shear stress on the survival and anti-chemotherapy ability of suspended circulating tumor cells during metastasis,and the effect of actomyosin activity on this regulation.In this study,we developed a circulatory system to generate physiologic levels of hemodynamic shear stress,which mimicked certain important aspects of the CTC microenvironment in blood circulation.The survival of tumor cells in suspension,as a model for real CTCs,under different shear stress and circulation duration was examined.We found that the majority of breast tumor cells s in suspension can be eliminated by hemodynamic shear stress.The surviving cells exhibit unique biophysical properties,including significantly retarded cell adhesion,mesenchymal-like cell morphology,and reduced F-actin expression and cellular stiffness.Cancer stem cells which has been reported in other papers have lower stiffness compared with conventional tumor cells showed significantly higher survival in blood flow.Importantly,low actomyosin activity promotes the survi 展开更多

This work investigates the machining temperatures of ultra-fine-grained titanium(UFG Ti),prepared by equal channel angular extrusion,through analytical modeling.UFG Ti has great usefulness in biomedical applications because of its high mechanical strength,sufficient manufacturability,and high biocompatibility.The temperatures were predicted using a physics-based predictive model based on material constitutive relation and mechanics of the orthogonal cutting process.The minimization between the stress calculated using Johnson–Cook constitutive model and the same stress calculated using mechanics model yields the estimation of machining temperatures at two deformation zones.Good agreements are observed upon validation to the values reported in the literature.The machinability of UFG Ti is investigated by comparing its machining temperature to that of Ti–6Al–4V alloy under the same cutting conditions.Significantly lower temperatures are observed in machining UFG Ti.The computational efficiency of the presented model is investigated by comparing its average computational time(~0.5 s)to that of a widely used modified chip formation model(8900 s)with comparable prediction accuracy.This work extends the applicability of the presented temperature model to a broader class of materials,specifically ultra-fine-grained metals.The high computational efficiency allows the in situ temperature prediction and optimization of temperature condition with process parameters planning. 展开更多

Supporting characteristics analysis of constant resistance bolts under coupled static-dynamic loading

To study the tensile mechanical properties of constant resistance bolts, the RFPA(Rock Failure Process Analysis) statics software is used to perform a uniaxial tensile test on a constant resistance bolt. The numerical test results show that the plastic strain value is 12 times the magnitude of the elastic strain. During plastic deformation, the fluctuation in the stress magnitude is relatively stable, indicating that the bolt has good constant resistance characteristics. The numerical test results are in good agreement with the laboratory test results of M.C. He, and the accuracy and reliability of the numerical test method are verified. Therefore, the RFPA software with coupled static-dynamic loading is further adopted to study the supporting effects of traditional bolts and constant resistance bolts under coupled staticdynamic loading. The numerical comparison of the test results show that the constant resistance bolts can effectively control the deformation amount and rate of the laneway surrounding rock, reduce the total and rate of increase in the accumulated acoustic emissions,decrease the stress on the units in the model and protect the stability of the laneway. This paper verifies that a constant resistance bolt has better impact resistance mechanical properties than those of a traditional bolt and provides an effective way to control rock burst and soft rock that is prone to large deformation damage. 展开更多

针对当前含孔边裂纹结构弹性断裂力学研究方法存在的研究结果与实际结果吻合性差、研究结果用户满意度低的问题,提出基于复变函数的含孔边裂纹结构弹性断裂力学问题研究方法。基于裂纹受力条件以及受力状况,将含孔边裂纹划分成两类:Ⅰ... 针对当前含孔边裂纹结构弹性断裂力学研究方法存在的研究结果与实际结果吻合性差、研究结果用户满意度低的问题,提出基于复变函数的含孔边裂纹结构弹性断裂力学问题研究方法。基于裂纹受力条件以及受力状况,将含孔边裂纹划分成两类:Ⅰ型裂纹和Ⅱ型裂纹。发现Ⅰ型裂纹的上下表面是相对张开的,Ⅱ型裂纹的上下表面沿横轴相对滑开。根据含孔边裂纹分析情况,利用复变函数法对含孔边裂纹结构弹性断裂力学问题予以解决,通过构建保角映射函数,并将无限放大的含孔边裂纹外部区域映射至单位圆内部或者是上半面,以此解决裂纹结构平面弹性相关问题,得到裂纹应力强度因子,实现含孔边裂纹结构弹性断裂力学研究。实验结果表明,基于复变函数的含孔边裂纹结构弹性断裂力学研究方法分析结果与实际情况吻合度最高为99%,研究结果用户满意度相较当前方法更高。所提方法整体性能优越,可为该领域研究提供一定的借鉴。 展开更多

角膜内皮细胞(CECs)对于维持角膜透明至关重要。成年人的CECs在体内不可进行有丝分裂,损伤后不可再生。体外培养的人CECs可进行有丝分裂,但是体外培养的人CECs传代后形态易趋向于成纤维细胞改变且标志性蛋白表达减少,发生角膜内皮细胞... 角膜内皮细胞(CECs)对于维持角膜透明至关重要。成年人的CECs在体内不可进行有丝分裂,损伤后不可再生。体外培养的人CECs可进行有丝分裂,但是体外培养的人CECs传代后形态易趋向于成纤维细胞改变且标志性蛋白表达减少,发生角膜内皮细胞间质转化(EndMT),成为体外培养CECs发展角膜再生医学的阻碍。同时,体内EndMT也参与到多种角膜病理改变,造成视力下降,甚至致盲,但是其发生机制尚未完全阐明。目前研究发现,CECs EndMT的机制可能涉及转化生长因子-β(TGF-β)信号通路、成纤维细胞生长因子(FGF)家族细胞因子、Notch信号通路。因此,相应的抑制角膜EndMT的方法有抑制TGF-β信号通路、使用PI3K抑制剂、应用小干扰RNA(siRNA)干扰p120、使用富含亮氨酸重复序列G蛋白偶联受体5(LGR5)及其配体、使用Notch信号传导抑制剂、敲低connexin43、使用基质金属蛋白酶抑制剂以及使用抗坏血酸-2-磷酸(Asc-2P)。本文就角膜EndMT的发生机制、间质转化抑制或逆转方法的研究进行综述。 展开更多

In this paper,stability properties of Tetrosomus gibbosus have been experimentally and numerically studied in pitching movements.This fish lives in southern offshores of Iran.Boxfishes usually live in extremely turbulent parts of seas and oceans,with high stability and maneuverability that has caught the attention of scientists.Their body shape let them swim with a quite rapid speed.Studies on Boxfishes have had influences on automotive and marine industries.The CAD file of the boxfish was created using optical CMM.Also a model was built and tested in a subsonic wind tunnel and a numerical study was conducted in virtual wind tunnel.The experimental results are consistent with the numerical ones and also support the findings of other studies on similar boxfishes.These results show that the flow around the fish,which is a consequence of its body shape,tries to maintain the stable position of the fish by resisting against the external force tilting the fish from its stable horizontal posture.The ventral and dorsal keels of the boxfish generate column-like vortices in horizontal direction which play an imperative role in maintaining the stability of the fish. 展开更多
