Involvement of sulfur assimilation in the low β subunit content of soybean seed storage protein revealed by comparative transcriptome analysis 预览

Merr.] seed storage protein is of great significance in sulfur-containing amino acid balance and soybean processing properties. The objective of this study was to elucidate the relationship between the β subunit and sulfur-containing amino acid composition, and the potential regulatory mechanism. The β subunit was independently accumulated in comparison with other major subunits (α/α′, acidic, basic, and A3) during seed filling, and a low level of β subunit content (BSC) was formed during the accumulation process. In low-BSC mature seeds, crude protein, oil content, and fatty acid composition were not changed, but sulfur-containing amino acids (Cys + Met) in the low- BSC seeds increased significantly (by 31.5%), suggesting that an internal regulatory mechanism within seed might be responsible for the rebalance of seed protein composition and that sulfur assimilation might be deeply involved in β subunit accumulation. Transcriptomic analysis revealed that genes involved in anabolism of cysteine, methionine, and glutathione were up-regulated but those involved in the catabolism of these compounds were down-regulated, suggesting a relationship between the elevation of methionine and glutathione and low BSC. Our study sheds light on seed composition in low BSC lines and on the potential molecular regulatory mechanism of β subunit accumulation, broadening our understanding of soybean seed protein synthesis and its regulation. 展开更多

目的探讨会阴部亚单位缺损的修复重建方案。方法对2008年1月至2018年12月解放军总医院第四医学中心烧伤整形五病区收治的21例会阴部缺损患者的临床资料进行回顾性分析。其中男10例,女11例,年龄4~68岁,平均26.4岁。受伤原因包括烧伤11例... 目的探讨会阴部亚单位缺损的修复重建方案。方法对2008年1月至2018年12月解放军总医院第四医学中心烧伤整形五病区收治的21例会阴部缺损患者的临床资料进行回顾性分析。其中男10例,女11例,年龄4~68岁,平均26.4岁。受伤原因包括烧伤11例,外伤2例,Paget病2例,Brown病2例,会阴部鳞癌3例,血管瘤1例。根据会阴部解剖结构,分为4个亚单位:1个前区(阴阜或耻骨联合)、2个中区(阴唇或阴囊)和1个后区(肛周)。会阴部病变切除后遗留创面23 cm×11 cm~5 cm×3 cm,根据会阴分区和组织缺损的情况选择合适的皮瓣进行修复。结果 21例患者中采用腹壁浅动脉岛状皮瓣修复9例,旋髂浅动脉皮瓣修复3例,腹壁浅动脉岛状皮瓣和旋髂浅动脉岛状皮瓣联合修复2例,阴部内动脉穿支皮瓣修复5例,股前外侧穿支皮瓣修复2例,皮瓣切取范围25 cm×12 cm~6 cm×3 cm,术后皮瓣全部成活,切口一期愈合。术后随访6个月至9年,平均13个月,供、受区外观良好,瘢痕隐蔽,患者满意。结论根据会阴部亚单位分区原则合理选择皮瓣对会阴部缺损进行修复重建,可更好地恢复其功能和外观,取得满意的临床效果。 展开更多

Nonmetal elements strictly govern the electrochemical performance of molybdenum compounds.Yet,the exact role played by nonmetals during electrocatalysis remains largely obscure.With intermetallic MoSi2comprising silicene subunits,we present an unprecedented hydrogen evolution reaction(HER)behavior in aqueous alkaline solution.Under continuous operation,the HER activity of MoSi2shows a more than one order of magnitude improvement in current density from 1.1 to 21.5 mA cmà2at 0.4 V overpotential.Meanwhile,this activation behavior is highly reversible,such that voltage withdrawal leads to catalyst inactivation but another operation causes reactivation.Thus,the system shows dynamics strikingly analogous to the legendary Sisyphus’labor,which drops and recovers in a stepwise manner repeatedly,but never succeeds in reaching the top of the mountain.Isomorphic WSi2behaves almost the same as MoSi2,whereas other metal silicides with silicyne subunits,including CrSi2and TaSi2,do not exhibit any anomalous behavior.A thin amorphous shell of MoSi2is observed after reaction,within which the Si remains partially oxidized while the oxidation state of Mo is basically unchanged.First-principles calculations further reveal that the adsorption of hydroxide ions on silicene subunit edges and the subsequent Si vacancy formation in MoSi2jointly lead to the anomalous HER kinetics of the adjacent Mo active centers.This work demonstrates that the role of nonmetal varies dramatically with the electronic and crystallographic structures of silicides and that silicene structural subunit may serve as a promoter for boosting HER in alkaline media. 展开更多

Objective: To explore genes potentially co-expressed with cyclin E in gastric cancer and discover possible targets for gastric cancer treatment. Methods: The Cancer Genome Atlas (TCGA) stomach adenocarcinoma sequencing data were used to predict genes co-expressed with cyclin E. Co-expression genes predicted by cBioPortal online analysis with Pearson correlation coefficient ≥0.4 were analyzed by gene ontology (GO) enrichment annotation using the PANTHER online platform (Ver. 7). Interactions between proteins encoded by these genes were analyzed using the STRING online platform (Ver. 10.5) and Cytoscape software (Ver. 3.5.1). Genes displaying a high degree of connection were analyzed by transcription factor enrichment prediction using FunRich software (Ver. 3). The significant transcription factor and cyclin E expression levels and their impact on gastric cancer progression were analyzed by Western blotting and Kaplan-Meier survival curve analysis. Results: After filtering the co-expression gene prediction results, 78 predicted genes that included 73 protein coding genes and 5 non-coding genes with Pearson correlation coefficient ≥0.4 were selected. The expressions of the genes were considered to be correlated with cyclin E expression. Among the 78 genes co-expressed with cyclin E, 19 genes at the central of the regulatory network associated with cyclin E were discovered. Nuclear transcription factor Y subunit alpha (NF-YA) was identified as a significant transcription factor associated with cyclin E co-expressing genes. Analysis of specimen donors’ clinical records revealed that high expression of NF-YA tended to be associated with increased cyclin E expression. The expression of both was associated with progression of gastric cancer. Western blotting results showed that compared with normal tissues, NF-YA and cyclin E were highly expressed in tumor tissues (P < 0.001). N-methyl-D-aspartate receptor hypofunction is the basis of pathophysiology in schizophrenia. Blocking the N-methyl-D-aspartate receptor impairs learning and memory abilities and induces pathological changes in the brain. Previous studies have paid little attention to the role of the N-methyl-D-aspartate receptor subunit 1 (NR1) in neurogenesis in the hippocampus of schizophrenia. A mouse model of schizophrenia was established by intraperitoneal injection of 0.6 mg/kg MK-801, once a day, for 14 days. In N-methyl-D-aspartate-treated mice, N-methyl-D-aspartate was administered by intracerebroventricular injection in schizophrenia mice on day 15. The number of NR1-, Ki67- or BrdU-immunoreactive cells in the dentate gyrus was measured by immunofluorescence staining. Our data showed the number of NR1-immunoreactive cells increased along with the decreasing numbers of BrdU- and Ki67-immunoreactive cells in the schizophrenia groups compared with the control group. N-methyl-D-aspartate could reverse the above changes. These results indicated that NR1 can regulate neurogenesis in the hippocampal dentate gyrus of schizophrenia mice, supporting NR1 as a promising therapeutic target in the treatment of schizophrenia. [Methods] Sixty Wistar rats were randomly divided into model group,Tiaomaiyin prescription group( whole prescription group),main efficacy group of removing heat to cool blood( blood cooling group),and auxiliary drug efficacy group of benefiting qi and nourishing heart( qi benefiting group),auxiliary efficacy group of promoting flow of qi and blood circulation( qi flow promoting group),and amiodarone group( western medicine group). Aconitine was given 7 d after the intragastric administration of the corresponding drugs,and the time of occurrence of arrhythmia in each group was observed. The left ventricular myocardium was subjected to reverse transcription-polymerase chain reaction and Western blotting. [Results] The ventricular premature beats( VPB) time in the whole prescription group and western medicine group was significantly longer than that in the model group. Ventricular tachycardia( VT),ventricular fibrillation( VF),and cardiac arrest( CA) were longer in the whole prescription group,blood cooling group,and western medicine group. The mRNA and protein expression of L-type calcium channel β2 subunit in the whole prescription group,blood cooling group and western medicine group were significantly decreased. [Conclusions] Tiaomaiyin whole prescription group and blood cooling group can reduce the occurrence time of tachyarrhythmia and reduce the expression of LTCC β2 in myocardium. 展开更多

为给黄淮南片冬麦区小麦育种提供种质信息,将1328份CIMMYT国际面包小麦观察圃(IBWSN)的种质于2011-2015年度种植于黄淮南片冬麦区的新乡,调查了42^nd~46^th国际面包小麦观察圃种质的主要农艺性状和抗病性,分析了45^th国际面包小麦观察... 为给黄淮南片冬麦区小麦育种提供种质信息,将1328份CIMMYT国际面包小麦观察圃(IBWSN)的种质于2011-2015年度种植于黄淮南片冬麦区的新乡,调查了42^nd~46^th国际面包小麦观察圃种质的主要农艺性状和抗病性,分析了45^th国际面包小麦观察圃种质的品质表现及优质亚基基因状况。结果表明,CIMMYT国际面包小麦观察圃种质均表现为春性,综合农艺性状差;株高不低于80cm,大多数(84.6%)在90~110cm之间;千粒重高于45.0g和50.0g的比例分别为28.4%和10.0%。CIMMYT国际面包小麦观察圃种质的花期与黄淮南片冬麦区品种一致,多数小麦种质白粉病发生比较轻。45^th国际面包小麦观察圃种质的湿面筋含量为23.6%~41.6%,其中,13.2%的小麦种质湿面筋含量大于35.0%,40.8%的小麦种质湿面筋指数不低于80.0%;49.7%的小麦种质携带Dx5+Dy10基因,3.0%的小麦种质携带Bx7OE基因;有10个小麦种质含有优质5+10亚基及7超量表达亚基,可作为优质亲本改良黄淮南片冬麦区高产品种。 展开更多

为评估包虫病基因工程亚单位疫苗的安全性和免疫效果,选取承德市接坝地区4个牧场479只8周龄羔羊进行免疫试验。免疫组(308只)于首次免疫前、加强免疫前,以及首次免疫后2、6和9个月,对照组(171只)于试验前和试验后9个月,采集羔羊血清,采... 为评估包虫病基因工程亚单位疫苗的安全性和免疫效果,选取承德市接坝地区4个牧场479只8周龄羔羊进行免疫试验。免疫组(308只)于首次免疫前、加强免疫前,以及首次免疫后2、6和9个月,对照组(171只)于试验前和试验后9个月,采集羔羊血清,采用ELISA抗体检测试剂盒进行抗体检测。结果显示:免疫疫苗后,个别羊有轻微的免疫反应;免疫组免疫前抗体阳性率为2.27%,加强免疫前抗体阳性率为87.16%,首次免疫后2、6和9个月的抗体阳性率分别为99.32%、93.49%和86.46%;对照组试验前抗体阳性率为2.34%,9个月后抗体阳性率为7.93%。结果表明,包虫病基因工程亚单位疫苗能够快速诱导机体产生抗体,并且抗体维持期较长,9个月后,动物个体仍维持较高的抗体水平。因此,该亚单位疫苗免疫效果及安全性较好,可以在包虫病流行区推广应用。 展开更多

The composition of high molecular weight glutenin subunits(HMW-GS)was analyzed using SDS-PAGE with 70 Longdong dryland winter wheat germplasm resources as experimental materials.The results showed that Longdong dryland winter wheat germplasm resources had N,7+8,2+12 as the dominant HMW-GS composition(42.86%),while the dominant HMW-GS composition in the introduced germplasms was N,7+9,2+12(36.84%).On the Glu-B1 locus of Longdong dryland germplasms,subunit 7+8 had the highest frequency,accounting for 65.71%,and on the Glu-B1 locus of the introduced germplasms,subunit 7+9 had the highest frequency,accounting for 57.89%.Whether subunit 7+8 is related to drought resistance in varieties still needs further study.On Glu-D1 locus,high-quality subunit 5+10 appeared in the introduced resources for 5 times,accounting for 13.16%of all the introduced lines,and appeared in Longdong dryland winter wheat germplasm resources for 2 times,only accounting for 2.86%of all the resources,suggesting that Longdong dryland germplasms lack high-quality subunits and need further improvement. 展开更多

The Brassicaceae species Braya humilis shows broad adaptation to different climatic zones and latitudes.However,the molecular adaptation mechanism of B.humilis is poorly understood.In China,B.humilis is mainly distributed on the Qinghai-Tibetan Plateau(QTP)and in the adjacent arid region.Previous transcriptome analysis of B.humilis has revealed that 39 salt and osmotic stress response genes are subjected to purifying selection during its speciation.To further explore the adaptation mechanism of B.humilis to an arid environment,OrthoMCL program was employed in this study and 6,268 pairs of orthologous gene pairs with high confidence were obtained between B.humilis and Arabidopsis thaliana.A comparative evolutionary analysis based on nonsynonymous to synonymous substitution ratio(Ka/Ks)was then conducted.There were 64 pairs exhibiting a Ka/Ks ratio more than 0.5 and among which,three instrumental candidate genes,T2_20487,T2_22576,and T2_13757,were identified with strong selection signatures(Ka/Ks>1).The corresponding A.thaliana orthologs are double-stranded RNA-binding domain protein,MADS-box family protein,and NADH-dehydrogenase subunit 6,which is encoded by mitochondria genome.This report not only demonstrates the adaptation contribution of fast evolving nuclear genes,but also highlights the potential adaptive value of mitochondria gene to the speciation and adaptation of B.humilis toward the extreme environment in an arid region. 展开更多

In China, it was first reported from Hainan Province, and later from several other provinces. In the present study, a rice root-knot nematode population found from the rice cultivation areas of Zhejiang Province, China is characterized via molecular analysis using internal transcribed spacer(ITS) and cytochrome c oxidase subunit Ⅱ(coxⅡ)-16 S rRNA genes and scanning electron microscopy(SEM) observations of males and the second-stage juveniles. Morphometric data and molecular sequence comparisons for all M. graminicola populations occurring in China are also provided. The overall morphology of M. graminicola found in Zhejiang match well with the original description, though males have a slightly longer body and stylet, and a shorter tail, while the second-stage juvenile is also slightly longer than in the original description. This is the first report of M. graminicola from Zhejiang. Phylogenetic studies based on coxⅡ suggest that all the Chinese populations belong to Type B. This study expands knowledge of the increasing distribution and phylogenetic relationships of M. graminicola that occur in China. 展开更多

【目的】探索不同萌发时期绿豆分离蛋白、清蛋白、球蛋白、谷蛋白和醇溶蛋白含量的动态变化规律,以及蛋白质组分(分离蛋白、球蛋白和清蛋白)的亚基组成及含量变化,为绿豆蛋白的加工利用提供科学依据。【方法】采用等电点沉淀法提取绿... 【目的】探索不同萌发时期绿豆分离蛋白、清蛋白、球蛋白、谷蛋白和醇溶蛋白含量的动态变化规律,以及蛋白质组分(分离蛋白、球蛋白和清蛋白)的亚基组成及含量变化,为绿豆蛋白的加工利用提供科学依据。【方法】采用等电点沉淀法提取绿豆分离蛋白,并根据Osborne分类法制备绿豆清蛋白、谷蛋白、球蛋白和醇溶蛋白,比较分析萌发前后绿豆分离蛋白及各蛋白组分含量的变化,同时通过SDS-PAGE电泳进一步分析萌发前后蛋白亚基组成及数量的变化。【结果】随着萌发时间的延长,绿豆分离蛋白的含量呈现先增高后下降的趋势,萌发36 h时与未萌发的绿豆相比提高了9.4%。绿豆清蛋白含量随着萌发时间的延长呈逐渐下降的趋势,萌发84 h时含量最小,为20.47 mg·g^-1。球蛋白随着萌发时间的延长呈现先升高后下降的趋势,萌发48 h时其含量最大,且比未萌发时提高了3.47倍。萌发对绿豆醇溶蛋白的含量变化影响不大。绿豆谷蛋白含量在萌发12—36 h和48—72 h变化无明显差异,但相对于未萌发的绿豆仍有一定程度的提高。绿豆蛋白酶活性在萌发36 h后不断上升,变化相对较大,72 h时开始下降。SDS-PAGE电泳图及光密度扫描分析结果发现,绿豆分离蛋白主要由7条条带组成(Ⅰ—Ⅶ),萌发过程中各条带相对含量不断减少,随着萌发时间的延长,分子量在25—66 kD的条带含量逐渐降低,分子量在18—25 kD的亚基条带含量在萌发的前72 h有所增加,萌发至96 h时几乎只剩下Ⅳ条带。绿豆清蛋白主要由4条条带组成(Ⅰ—Ⅳ),分子量分别为61.56、48.99、29.88和20.42 kD,萌发过程中条带Ⅰ相对含量从0 h的18.4%降至60 h的16.4%,萌发72 h后条带Ⅰ消失;条带Ⅱ在萌发过程中始终存在,但是含量不断减少;条带Ⅲ和条带Ⅳ也在萌发过程中不断减少,萌发72 h后消失。同时,分子量为18—25 kD的亚基条带相对含量� 展开更多
