SLC38A1 Promotes Proliferation and Migration of Human Colorectal Cancer Cells 预览

Current studies have demonstrated that SLC38A1 proteins play a causal role in neoplastic cell transformation. The twofold aim of this study was to provide insight into whether a variance in the expression of SLC38A1 exists between human colorectal cancer and healthy human tissues and to determine how silencing or overexpressing the SLC38A1 gene could affect the proliferation, viability and migration of colorectal cancer cells. Immunohistochemical staining was used to analyze the expression of SLC38A1 in colorectal cancer tissues and adjacent normal mucosa in 77 patients who underwent surgical resection. The expression of SLC38A1 in colorectal cancer tissues and cell lines was detected using RT-PCR and Western blotting. Two colorectal cancer cell lines SW480 and HCT116 were used to examine whether silencing SLC38A1 with si RNA and overexpressing SLC38A1 with sh RNA could affect cell viability and migration. As a result, the SLC38A1 protein was very low or undetectable in the normal colon mucosa. In contrast, strong staining of SLC38A1 protein was found in the cytoplasm in 79.2% colorectal cancer samples. More pronounced SLC38A1 expression in colorectal cancer tissues was significantly associated with tumor node metastasis(TNM) stage. Inhibition of SLC38A1 reduced tumour growth and suppressed proliferation and migration of SW480 cells. In contrast, overexpression of SLC38A1 had the opposite effects on HCT116 cells. SLC38A1 is overexpressed in colorectal cancer, which suggests that it is associated with tumour progression. These results encourage the exploration of SLC38A1 as a target for intervention in colorectal cancer. 展开更多

Essential Oil from Siegesbeckia pubescens Induces Apoptosis through the Mitochondrial Pathway in Human HepG2 Cells 预览

Siegesbeckia pubescens(SP) has been used as a traditional medicine for the treatment of and inflammatory diseases. However, the activities of SP against hepatocellular carcinoma and the related mechanisms remain unclear. The present study aimed to examine the effects of the essential oil of SP(SPEO) on the proliferation of hepatocellular carcinoma cells and the possible mechanisms. The growth inhibition of Hep G2 cells was analyzed by MTT assay. Hoechst 33258 and fluorescence microscopy were utilized to observe the nuclear morphological changes of apoptotic cells. Flow cytometry was used to detect cell apoptosis and cell cycle. The expressions of the target proteins were detected by Western blotting. The results showed that SPEO obviously inhibited the proliferation of Hep G2 cells in a dose-dependent manner. SPEO activated a series of apoptotic proteins in Hep G2 cells, increasing expression levels of Bax, caspase-3 and caspase-9, and decreasing the bcl-2 expression level. SPEO displayed promising anti-hepatocellular carcinoma activities in vitro, partly by inducing apoptosis in Hep G2 cells through activating the mitochondrial pathway. 展开更多

Bisphenol-A(BPA) has been considered as an endocrine disrupting chemical(EDC) because it can exert estrogenic properties.For bisphenol-S(BPS) and bisphenol-F(BPF) that are BPA analogs and substitutes,their risk to estrogendependent cancer has been reported rarely compared with the numerous cases of BPA.In this study,we examined whether BPA,BPS,and BPF can lead to the proliferation,migration,and epithelial mesenchymal transition(EMT) of MCF-7 clonal variant(MCF-7 CV) breast cancer cells expressing estrogen receptors(ERs).In a cell viability assay,BPA,BPS,and BPF significantly increased proliferation of MCF-7 CV cells compared to control(DMSO) as did17β-estradiol(E2).In Western blotting assay,BPA,BPS,and BPF enhanced the protein expression of cell cycle progression genes such as cyclin D1 and E1.In addition,MCF-7 CV cells lost cell to cell contacts and acquired fibroblast-like morphology by the treatment of BPA,BPS,or BPF for 24 hours.In cell migration assay,BPA,BPS,and BPF accelerated the migration capability of MCF-7 CV cells as did E2.In relation with the EMT process,BPA,BPS,and BPF increased the protein expression of N-cadherin,while they decreased the protein expression of Ecadherin.When BPA,BPS,and BPF were co-treated with ICI 182,780,an ER antagonist,proliferation effects were reversed,the expression of cyclin D1 and cyclin E1 was downregulated,and the altered cell migration and expression of N-cadherin and E-cadherin by BPA,BPS,and BPF were restored to the control level.Thus,these results imply that BPS and BPF also have the risk of breast cancer progression as much as BPA in the induction of proliferation and migration of MCF-7 CV cells by regulating the protein expression of cell cycle-related genes and EMT markers via the ER-dependent pathway. 展开更多

目的:探讨磷酸酶–张力蛋白基因(phosphatase and tensin homology deleted on chromosome ten,PTEN)在正常脑组织及星形细胞瘤中的表达及其意义。方法:采用免疫组织化学法及RT-PCR方法检测正常脑组织及不同分级星形细胞瘤组织中PTE... 目的:探讨磷酸酶–张力蛋白基因(phosphatase and tensin homology deleted on chromosome ten,PTEN)在正常脑组织及星形细胞瘤中的表达及其意义。方法:采用免疫组织化学法及RT-PCR方法检测正常脑组织及不同分级星形细胞瘤组织中PTEN蛋白及mRNA的表达水平,探讨并分析其与星形细胞瘤组织学分级的关系,Western印迹法检测正常脑组织及星形细胞瘤脑脊液中PTEN蛋白的表达。结果:免疫组织化学方法显示PTEN在正常脑组织中阳性率较高,为90%(9/10),星形细胞瘤中阳性率较低,为47.5%(19/40),且PTEN在不同的组织学分级中,阳性率不同。RT-PCR结果显示PTEN m RNA在正常脑组织中表达较高,其相对表达量为0.861±0.072,显著高于星形细胞瘤组织中的表达(0.127±0.008),差异有统计学意义(P〈0.05),并随着组织学级别增加,其表达呈降低趋势。Western印迹法结果显示P TEN蛋白在非星形细胞瘤组脑脊液中的相对表达量为1.549±0.194,高于星形细胞瘤组脑脊液中的相对表达量(0.602±0.058),差异具有统计学意义(P〈0.05)。结论:检测PTEN基因蛋白表达有望作为评估人星形细胞瘤生物学行为和预后的参考指标。 展开更多

目的:观察顺铂诱导急性肾损伤(acute kidney injury,AKI)模型中肾皮质Persephin(PSP)的变化。方法:雄性C57BL/6小鼠随机分为对照组及模型组,模型组应用顺铂单次腹腔注射(15mg/kg)制备AKI小鼠模型,对照组小鼠予以同等容积的0.9%... 目的:观察顺铂诱导急性肾损伤(acute kidney injury,AKI)模型中肾皮质Persephin(PSP)的变化。方法:雄性C57BL/6小鼠随机分为对照组及模型组,模型组应用顺铂单次腹腔注射(15mg/kg)制备AKI小鼠模型,对照组小鼠予以同等容积的0.9%的生理盐水单次腹腔注射,造模3天后观察各组小鼠肾功能变化、肾脏病理学改变、实时PCR法及Western印记法检测肾脏皮质PSP的表达。结果 :与对照组相比,模型组小鼠出现精神萎靡不振,反应迟钝,活动减少,造模后第3天尿素氮明显上升(P〈0.05),肾脏病理改变中模型组肾小管管型计数(0.63±0.62个/HPF)及坏死计数(3.71±1.14个/HPF)均高于正常对照组(P〈0.05)。PSP在模型组肾皮质的表达高于对照组(P〈0.05)。结论:AKI模型中肾脏皮质PSP表达升高,提示PSP可能参与了AKI的病程,但其相关作用机制尚需进一步研究。 展开更多

IFN-γ Secretion in Gut of Ob/Ob Mice after Vertical Sleeve Gastrectomy and Its Function in Weight Loss Mechanism 预览

This study aimed to test interferon(IFN)-... Vertical sleeve gastrectomy(VSG) is becoming more and more popular among the world. Despite its dramatic efficacy, however, the mechanism of VSG remains largely undetermined. This study aimed to test interferon(IFN)-γ secretion n of mesenteric lymph nodes in obese mice(ob/ob mice), a model of VSG, and its relationship with farnesoid X receptor(FXR) expression in the liver and small intestine, and to investigate the weight loss mechanism of VSG. The wild type(WT) mice and ob/ob mice were divided into four groups: A(WT+Sham), B(WT+VSG), C(ob/ob+Sham), and D(ob/ob+VSG). Body weight values were monitored. The IFN-γ expression in mesenteric lymph nodes of ob/ob mice pre- and post-operation was detected by flow cytometry(FCM). The FXR expression in the liver and small intestine was detected by Western blotting. The mouse AML-12 liver cells were stimulated with IFN-γ at different concentrations in vitro. The changes of FXR expression were also examined. The results showed that the body weight of ob/ob mice was significantly declined from(40.6±2.7) g to(27.5±3.8) g on the 30 th day after VSG(P<0.05). At the same time, VSG induced a higher level secretion of IFN-γ in mesenteric lymph nodes of ob/ob mice than that pre-operation(P<0.05). The FXR expression levels in the liver and small intestine after VSG were respectively 0.97±0.07 and 0.84±0.07 fold of GAPDH, which were significantly higher than pre-operative levels of 0.50±0.06 and 0.48±0.06 respectively(P<0.05). After the stimulation of AML-12 liver cells in vitro by different concentrations of IFN-γ(0, 10, 25, 50, 100, and 200 ng/m L), the relative FXR expression levels were 0.22±0.04, 0.31±0.04, 0.39±0.05, 0.38±0.05, 0.56±0.06, and 0.35±0.05, respectively, suggesting IFN-γ could distinctly promote the FXR expression in a dose-dependent manner in comparison to those cells without IFN-γ stimulation(P<0.05). OBJECTIVE: To investigate the effect of Gubenfangxiao decoction(GBFXD) on respiratory-syncytial-virus(RSV)-induced asthma and the expression of asthma susceptibility gene, orosomucoid 1-like protein 3(ORMDL3) in mice.METHODS: Seventy-two female BALB/c mice were randomly assigned to normal, model, GBFXD high dose, GBFXD moderate dose, GBFXD low dose and montelukast groups. An asthma model was induced via intraperitoneal injection and aerosol inhalation of ovalbumin(OVA) and repeated intranasal instillation of RSV in all mice, except those in the normal group. All treatments were administered at the first onset of asthma(within 8 weeks of model establishment) and the mice were euthanized after 28 days of treatment. The levels of transforming growth factor-β(TGF-β) and interleukin-6(IL-6) in bronchoalveolar lavage fluid(BALF) of the mice were measured and the expression of asthma susceptibility gene ORMDL3 in lung tissue was determined using real-time polymerase chain reaction(RT-PCR) and western blotting.RESULTS: Expression of ORMDL3 and levels of TGF-β and IL-6 were significantly higher in the model group(P < 0.05, P < 0.01) compared with the normal mice. Levels of ORMDL3, TGF-β and IL-6 were significantly lower in all three GBFXD treated groups(P < 0.05) compared with the model group.However, the levels in the GBFXD treatment groups did not differ significantly from the montelukast group.CONCLUSION: GBFXD had a therapeutic effect in this experimental model. The functional mechanism of GBFXD may involve multiple factors, including alleviation of airway inflammation, down-regulation of asthma susceptibility gene ORMDL3 and inhibition of airway remodeling. 展开更多

Mcl-1 as a Potential Therapeutic Target for Human Hepatocelluar Carcinoma 预览

Hepatocellular carcinoma(HCC) is a major cause of cancer-related mortality in part due to its high resistance to chemotherapeutic drugs. The anti-apoptotic Mcl-1 expression has been reported as a resistance factor in various types of tumors. Here, we investigated the expression of Mcl-1 in hepatoma cells and HCC tissues and its relationship with p53, and analyzed the possibility of the gene as a molecular target for HCC therapy. HCC specimens of 30 patients were examined by immunohistochemistry for Mcl-1 and p53 expression. Mcl-1 expression in hepatoma cell lines was measured by RT-PCR and Western blotting. The suppression of Mcl-1 by RNA interference or specific phosphatidylinositol-3 kinase(PI3K) inhibitor, LY294002, was evaluated as monotherapy, and it was combined with mitomycin C(MMC) in treating hepatoma cell line Hep G2. Cell viability and apoptosis were assessed by MTT and FACS analysis. Finally, changes of Mcl-1 or p53 expression in various hepatoma cell lines were examined after transfection with Mcl-1 si RNA, the Mcl-1 expression plasmid, or the wide-type p53 expression plasmid, respectively. Mcl-1 protein was remarkably enhanced in HCC tissues as compared with adjacent non-tumor liver tissues. In addition, Mcl-1 was prominently expressed in Hep G2 and Hep3 B cells, weakly in SMMC7721 cells, and not in L02 cells. P53 protein was also overexpressed in HCC tissues and there was a significant correlation between the expression of p53 and Mcl-1. Silencing Mcl-1 by RNAi or LY294002 downregulated Mcl-1 expression and led to decreased cell viability and increased apoptosis. Combination of MMC and Mcl-1 RNAi or LY294002 exhibited a significant chemosensitizing effect. The expression of p53 was not influenced by Mcl-1 si RNA in Hep G2 cells or transfection with the Mcl-1 expression plasmid in L02 cells. Furthermore, the expression of Mcl-1 in Hep3 B cells was also not significantly changed after transfection with the wild-type p53 expression plasmid.
