A new assay method for natural killer (NK) cell activity was established using quantitative RT-PCR (RT-qPCR) to determine the gene expression of granzyme B (GzmB) and perforin 1 (Prf1). The RT-qPCR method was compared to a conventional cytotoxic assay. Upregulated expression of GzmB and Prf1 mRNA and enhanced cytotoxic activity were observed in splenocytes from lipopolysaccharide (LPS)-treated rats. A high correlation, R<sup>2</sup> = 0.71, was observed between the gene expression of GzmB and the cytotoxic activity of splenocytes from these rats, indicating that GzmB RT-qPCR is a reliable alternative method to assess NK cell activity/activation. Remarkably, 12.6- to 59.7-fold upregulation of GzmB mRNA expression was observed in leukocytes, the spleen, and splenocytes from LPS-injected rats. Its upregulation appeared to be dose-dependent on the LPS concentration in the range of 0.01 - 0.1 mg/kg. Whereas, only 1.3- to 1.9-fold increase of cytotoxic activity was detected in splenocytes from the rats treated with LPS in the same range. From these, it is evident that, to assess NK cell activity/activation, the GzmB RT-qPCR method is highly sensitive compared with the conventional cytological assay. Furthermore, this GzmB RT-qPCR method is advantageous, as it does not require freshly prepared splenocytes and cell culture procedures. The convenience of GzmB RT-qPCR enables the use of whole blood, leukocytes, the spleen, and/or their frozen samples to evaluate NK cell activity/activation. 展开更多

Simulating the Production of Medical Radioisotopes in a Fast Thorium-Cycle ADS with SERPENT 认领

Radiopharmaceuticals are used in nuclear medicine for diagnostic or therapeutic acts. The short decay half-lives of medical radioisotopes, especially those used for diagnostics, imply that they should be produced continuously and transported as quickly as possible to the medical units where they are used. Neutron-rich medical radioisotopes are generally produced in research reactors, like technetium-99m, lutetium-177, holmium-166 and iodine-131. On the other hand, proton-rich radioisotopes are produced via reactions with charged particles from accelerators like fluorine-18, gallium-67, iodine-123 and thallium-201. Beside this, innovative nuclear reactors are advocated as solutions to the issues of nuclear waste production and proliferation threats. Fast neutron, thorium-cycle and accelerator-driven subcritical (ADS) reactors are some of the most promising of them, proposed as safer fuel breeders and “waste burners”. This article examines the use of a fast thorium-cycle ADS with liquid lead-bismuth eutectic coolant for the production of molybdenum-99/technetium-99m and lutetium-177. Burnup simulation has been made with the Monte-Carlo (MC) code SERPENT. It is demonstrated that MC codes can advantageously be used to determine the optimal irradiation time for a given radioisotope in a realistic reactor core. It is also shown that fast thorium-cycle ADS is an economical option for the production of medical radioisotopes. 展开更多

Adsorption and Kinetic Study of Activated Carbon Produced from Post-Consumer Low-Density Polyethylene (LDPE) Wastes 认领

Post-consumer polymeric wastes in form of low-density polyethylene (LDPE) can now be considered suitable as a precursor for the synthesis of low-cost activated carbon (AC). This study produced AC from LDPE using sulphuric acid (H<sub>2</sub>SO<sub>4</sub>) and potassium hydroxide (KOH) as the activating agent. The reaction conditions for pyrolysis were varied in the range of 0.50 - 2.00 M, 400<span style="color:#4F4F4F;font-family:-apple-system, " font-size:16px;white-space:normal;background-color:#ffffff;"="">°</span>C - 500<span style="color:#4F4F4F;font-family:-apple-system, " font-size:16px;white-space:normal;background-color:#ffffff;"="">°</span>C, and 45 - 60 minutes. Physico-chemical investigations reveal that AC yield is significantly dependent on both carbonization temperatures and time. The obtained optimum values of 446.50<span style="color:#4F4F4F;font-family:-apple-system, " font-size:16px;white-space:normal;background-color:#ffffff;"="">°</span>C and 51.09 mins gave a yield of 24% for the base-activated carbon. The high iodine numbers obtained strongly indicate the presence of large surface area and pore volumes is further confirmed using the Scanning Electron Microscopy (SEM) analysis which reveals the presence of pores on the external surface of the carbons. Fourier Transform Infrared Technique (FTIR) analysis further shows that the synthesized compounds are purely carbon with rich oxy-gen-surface complexes on the surface which is as a result of the introduction of the chemical oxidizing agents. The produced carbons were found to have high adsorption affinity for selected inorganic ions which are: Mn<sup>7+</sup>, Co<sup>2+</sup>, and Cr<sup>6+</sup>. Adsorption isotherm results show the adsorption process to be favourable with the Langmuir isotherm parameter RL having values of <1, while the Freudlich adsorption model was found to perfectly fit the data at selected adsorbent dosages and adsorbate concentrations. AIM:To investigate the changes of Iba-1 and other potential markers for microglia activation in experimental diabetic retinopathy(DR).METHODS:Male Sprague-Dawley rats were rendered diabetes via intraperitoneal injection of streptozotocin.The retinas were harvested at 1 to 24 wk after diabetes onset.Hypoxia-treated mouse microglial cell line(BV2 cells)was employed as the in vitro model to mimic diabetic condition.The expressions of Iba-1,CD11 b,ICAM-1 as well as the inflammatory factors were examined with real-time polymerase chain reaction,Western blot and immunofluorescence both in vivo and in vitro.RESULTS:Compared with age-matched normal control,the number of microglia(Iba-1 positive immunostaining)in diabetic rat retinas was increased from 1 to 24 wk of diabetes,which was most obvious at 12 wk of diabetes.Iba-1 protein expression detected by Western blot was increased slightly in diabetic rat retinas compared with that in age-matched normal control;however,there was statistically significant between two groups only at 2 wk after diabetes onset.The m RNA expression of Iba-1 was decreased significantly at 2 and 4 wk of diabetic rat retinas,and remained unchanged at 8 and 12 wk of diabetes.In BV2 cells,there was no significant change for the Iba-1 protein expression between normoxia and hypoxia groups;however,its m RNA level was decreased significantly under hypoxia.To further characterize microglial activation,F4/80,CD11 b and inflammatory factors were detected both in vivo and in vitro.Compared with normal control,the expressions of F4/80 and CD11 b as well as the inflammatory factors,such as ICAM-1,i NOS,COX2,IL-1βand IL-6,were increased significantly both in vivo and in vitro.CONCLUSION:Iba-1 protein expression might not be a sensitive marker to evaluate the activation of microglia in experimental DR.However,Iba-1 immunostaining,in combination with other markers like CD11 b and ICAM-1,could be well reflect the activation of microglia.Thus,it is of great importance to explore other potential marker to evaluate the activation of microglia. 展开更多

Through this strategy, a series of mononuclear, trinuclear and tetranuclear cyclometalated iridium complexes containing Cp*Ir—B or Cp*Ir—C bonds were successfully isolated in a high yield. Sperm-specific phospholipase C zeta(PLCζ)initiates intracellular calcium(Ca2+)transients which drive a series of concurrent events collectively termed oocyte activation.Numerous investigations have linked abrogation and absence/reduction of PLCζwith forms of male infertility in humans where oocyte activation fails.However,very few studies have examined potential relationships between PLCζand advancing male age,both of which are increasingly considered to be major effectors of male fertility.Initial efforts in humans may be hindered by inherent PLCζvariability within the human population,alongside a lack of sufficient controllable repeats.Herein,utilizing immunoblotting,immunofluorescence,and quantitative reverse transcription PCR(qRT-PCR)we examined for the first time PLCζprotein levels and localization patterns in sperm,and PLCζmRNA levels within testes,from mice at 8 weeks,12 weeks,24 weeks,and 36 weeks of age,from two separate strains of mice,C57BL/6(B6;inbred)and CD1(outbred).Collectively,advancing male age generally diminished levels and variability of PLCζprotein and mRNA in sperm and testes,respectively,when both strains were examined.Furthermore,advancing male age altered the predominant pattern of PLCζlocalization in mouse sperm,with younger mice exhibiting predominantly post-acrosomal,and older mice exhibiting both post-acrosomal and acrosomal populations of PLCζ.However,the specific pattern of such decline in levels of protein and mRNA was strain-specific.Collectively,our results demonstrate a negative relationship between advancing male age and PLCζlevels and localization patterns,indicating that aging male mice from different strains may serve as useful models to investigate PLCζin cases of male infertility and subfertility in humans. 展开更多

A great deal of attention has been paid on developing plant-derived hard carbon(HC)materials as anodes for sodium-ion batteries(SIBs).So far,the regulation of HC has been handicapped by the well-known ambiguity of Na^(+)storage mechanism,which fails to differentiate the Na^(+)adsorption and Na^(+)insertion,and their relationship with the size of d-interlayer spacing and structural porosity.Herein,bagassederived HC materials have been synthesized through a combination of pyrolysis treatment and microwave activation.The combined protocol has enabled to synergistically control the d-interlayer spacing and porosity.Specifically,the microwave activation has created slit pores into HC and these pores allow for an enhanced Na^(+)adsorption with an increased sloping capacity,establishing a strong correlation between the porosity and sloping capacity.Meanwhile,the pyrolysis treatment promotes the graphitization and it contributes to an intensified Na^(+)insertion with an increased plateau capacity,proving that the plateau capacity is largely contributed by the Na^(+)insertion between interlayers.Therefore,the structural regulation of bagasse-derived HC has provided a proof on positively explaining the Na^(+)storage with HC materials.The structural changes in the pore size distribution,specific surface area,d-interlayer spacing,and the electrochemical properties have been comprehensively characterized,all supporting our understanding of Na^(+)storage mechanism.As a result,the HC sample with an optimized d-interlayer spacing and porosity has delivered an improved reversible capacity of 323.6 m Ah g^(-1) at 50 m A g^(-1).This work provides an understanding of Na^(+)storage mechanism and insights on enhancing the sloping/plateau capacity by rationally regulating the graphitization and porosity of HC materials for advanced SIBs. 展开更多

Aluminum-air(Al-air) batteries are promising candidates for energy storage applications because of their high theoretical energy density and low cost.Nevertheless,their developments have been severely hindered by multiple obstacles,among which the activation and self-corrosion inhibition of Al anode have been considered to be significant challenges.In neutral electrolytes,the main problem is the activation of Al anode,while the self-corrosion of Al anode becomes dominant in alkaline electrolytes.Considerable efforts have been devoted to overcoming the dilemma associated with the Al anode.This review firstly underscores the underlying mechanisms of passivation and self-corrosion of Al anode in different electrolytes.Then,specific attentions are paid to Al alloy anode,including the role of various elements and standard processing technology.Finally,general conclusions,current limitations,and future perspectives on Al alloy anode are presented. 展开更多

以脱脂棉、天丝和剑麻为原料,KOH溶液为活化剂,通过炭化、活化制备活性炭纤维(ACFs),研究炭化温度和碱碳比对ACFs物理化学性质的影响,并借助比表面积测试(BET)、X射线衍射分析(XRD)等手段对纤维进行表征。结果表明,3种原料炭化致孔的难... 以脱脂棉、天丝和剑麻为原料,KOH溶液为活化剂,通过炭化、活化制备活性炭纤维(ACFs),研究炭化温度和碱碳比对ACFs物理化学性质的影响,并借助比表面积测试(BET)、X射线衍射分析(XRD)等手段对纤维进行表征。结果表明,3种原料炭化致孔的难易程度为:脱脂棉>剑麻>天丝。当炭化温度为700℃时,炭化材料的比表面积分别为390.4、48.8和179.2 m^(2)·g^(-1)。采用质量分数w为10%的KOH溶液活化,碱碳质量比为2:1,脱脂棉、剑麻基ACFs的比表面积分别为1375和1161 m^(2)·g^(-1)。采用w为20%的KOH溶液活化,当碱碳质量比为3:1时,天丝基ACFs的比表面积可达1594 m^(2)·g^(-1),孔径分布比前两者窄。 展开更多

The two-drug combined chemotherapy of platinum and fluorouracil has been reported to efficiently kill tumor cells as the first-line treatment for advanced gastric cancer.However,the effect of these drugs on T cells remains unclear.Here,we showed that T cells including CD4+T cells and CD8+T cells of the patients with advanced gastric cancer after platinum and fluorouracil chemotherapy exhibited enhanced ex vivo proliferation ability as compared to that before chemotherapy.In addition,platinum and fluorouracil also promoted the differentiation of human T cells into Th1 and Th9 subtypes and cytotoxic T lymphocytes(CTLs)in vitro and in vivo.Accordingly,the combination therapy greatly suppressed tumor growth with increased tumor infiltration of Th1,Th9,and CTL cells in a mouse tumor model.Moreover,in activated T cells,long-term treatment with these two drugs further facilitates T cell activation along with promoted nuclear factor-κB(NF-κB)activation.Our findings demonstrate a previously unidentified function of platinum and fluorouracil combination chemotherapy in promoting T cell–mediated antitumor immunity. 展开更多

Mice use ultrasonic vocalizations(USVs)to communicate each other and to convey their emotional state.USVs have been greatly characterized in specific life phases and contexts,such as mother isolation-induced USVs for pups or female-induced USVs for male mice during courtship.USVs can be acquired by means of specific tools and later analyzed on the base of both quantitative and qualitative parameters.Indeed,different ultrasonic call categories exist and have already been defined.The understanding of different calls meaning is still missing,and it will represent an essential step forward in the field of USVs.They have long been studied in the ethological context,but recently they emerged as a precious instrument to study pathologies characterized by deficits in communication,in particular neurodevelopmental disorders(NDDs),such as autism spectrum disorders.This review covers the topics of USVs characteristics in mice,contexts for USVs emission and factors that modulate their expression.A particular focus will be devoted to mouse USVs in the context of NDDs.Indeed,several NDDs murine models exist and an intense study of USVs is currently in progress,with the aim of both performing an early diagnosis and to find a pharmacological/behavioral intervention to improve patients’quality of life. 展开更多
