Rapid discovery and identification of the anti-inflammatory constituents in Zhi-Shi-Zhi-Zi-Chi-Tang

The anti-inflammatory active ingredients of Zhi-Shi-Zhi-Zi-Chi-Tang(ZSZZCT), a traditional Chinese medicine formula, were predicted and identified using an approach based on activity index, LC-MS, semi-preparative LC and NMR. Firstly, the whole extract of ZSZZCT was analyzed using liquid chromatography-quadrupole time of flight-mass spectrometry(LC-QTOF-MS) and liquid chromatography-ion trap mass spectrometry(LC-IT-MS), 79 constituents were detected and 39 constituents were identified unambiguously or tentatively. Subsequently, the whole extract of the formula was separated into multiple components and the activity index method was used to calculate index values of the 79 constituents by integrating the chemical and pharmacological information of multiple components. Four polymethoxyl flavones were predicted as the major active constituents according to the activity index values. Furthermore, three polymethoxyl flavones were prepared using the strategy with semipreparative LC guided by LC-MS, and their anti-inflammatory activities were validated. The results show that three polymethoxyl flavones with higher positive index values, i.e., 3, 5, 6, 7, 8, 3’, 4’-heptamethoxyflavone, 3-hydroxynobiletein and tangeretin had significant anti-inflammatory effects. In conclusion, the predicted results indicated that the activity index method is feasible for the accurate prediction of active constituents in TCM formulae. 展开更多

Osmundea pinnatifida is a red edible seaweed known as pepper dulse.O.pinnatifida was cultivated in the farm of ALGAplus(ílhavo,Portugal).This farm is integrated with a seabream and seabass commercial aquaculture and uses the nutrient-enriched water resultant from the fish production as its cultivation medium in the integrated multi-trophic aquaculture(IMTA)manner.Wild and IMTA-cultivated samples of O.pinnatifi da were screened for antioxidant and antitumor activities.The antioxidant capacity of solvent extracts from wild and IMTA cultivated samples was assessed in two methods(2,2-diphenyl-1-picrylhydrazyl(DPPH)and oxygen radical absorbance capacity(ORAC)),and their total phenolic contents(TPC)were estimated.Antitumor activity was evaluated in three different tumor cell lines(HepG-2,MCF-7,and SH-SY5Y)through 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay.Among the solvents used for extraction,dichloromethane was the most eff ective to extract phenolic compounds and presented higher ORAC.A significant correlation was found between TPC and ORAC,which was also sustained by the principal components analysis(PCA).Dichloromethane extracts induced a cytostatic eff ect on MCF-7 cells and showed weak cytotoxicity to SH-SY5Y cells and weak impact on cell proliferation.Overall,there were no statistically signifi cant differences in the biological activities shown by the wild and IMTA-cultivated samples.Hence,O.pinnatifida can be obtained in an economical and environmentally sustainable way through IMTA,maintaining bioactive properties in a high potential for further nutraceutical purposes. 展开更多

The antioxidant and anti-inflamma... Objective A strain of Aspergillus niger(A. niger), capable of releasing bound phenolic acids from wheat bran, was isolated. This strain was identified by gene sequence identification. The antioxidant and anti-inflammatory capacity of ferulic acid released from wheat bran by this A. niger strain(FA-WB) were evaluated. Methods Molecular identification techniques based on PCR analysis of specific genomic sequences were conducted;antioxidant ability was examined using oxygen radical absorbance capacity(ORAC), cellular antioxidant activity(CAA) assays, and erythrocyte hemolysis assays. RAW264.7 cells were used as a model to detect anti-inflammatory activity. Results The filamentous fungal isolate was identified to be A. niger. ORAC and CAA assay showed that FA-WB had better antioxidant activity than that of the ferulic acid standard. The erythrocyte hemolysis assay results suggested that FA-WB could attenuate AAPH-induced oxidative stress through inhibition of reactive oxy gen species(ROS) generation. FA-WB could significantly restore the AAPH-induced increase in intracellular antioxidant enzyme activities to normal levels as well as inhibit the intracellular malondialdehyde formation. TNF-?, IL-6, and NO levels indicated that FA-WB can inhibit the inflammation induced by lipopolysaccharide(LPS). Conclusion Ferulic acid released from wheat bran by a new strain of A. niger had good anti-inflammatory activity and better antioxidant ability than standard ferulic acid. 展开更多

Antimicrobial peptides(AMPs) are a group of gene-encoded small peptides that play pivotal roles in the host immune system of multicellular organisms.Cathelicidins are an important family of AMPs that exclusively exist in vertebrates. Many cathelicidins have been identified from mammals, birds, reptiles and fish. To date, however, cathelicidins from amphibians are poorly understood. In the present study, two novel cathelicidins(OL-CATH1 and 2) were identified and studied from the odorous frog Odorrana livida.Firstly, the cDNAs encoding the OL-CATHs(780 and735 bp in length, respectively) were successfully cloned from a lung cDNA library constructed for the frog. Multi-sequence alignment was carried out to analyze differences between the precursors of the OL-CATHs and other representative cathelicidins.Mature peptide sequences of OL-CATH1 and 2 were predicted(33 amino acid residues) and their secondary structures were determined(OL-CATH1 showed a random-coil conformation and OL-CATH2 demonstrated α-helical conformation). Furthermore,OL-CATH1 and 2 were chemically synthesized and their in vitro functions were determined. Antimicrobial and bacterial killing kinetic analyses indicated that OL-CATH2 demonstrated relatively moderate and rapid antimicrobial potency and exhibited strong anti-inflammatory activity. At very low concentrations(10 μg/mL), OL-CATH2 significantly inhibited the lipopolysaccharide(LPS)-induced transcription and production of pro-inflammatory cytokines TNF-α, IL-1βand IL-6 in mouse peritoneal macrophages. In contrast, OL-CATH1 did not exhibit any detectableantimicrobial or anti-inflammatory activities. Overall,identification of these OL-CATHs from O. livida enriches our understanding of the functions of cathelicidins in the amphibian immune system. The potent antimicrobial and anti-inflammatory activities of OL-CATH2 highlight its potential as a novel candidate in anti-infective drug development. 展开更多

Objective To assess the activities of biapenem against multidrug-resistant and extensively drug-resistant Mycobacterium tuberculosis. Methods Biapenem/clavulanate(BP/CL) was evaluated for in vitro activity against Mycobacterium tuberculosis(Mtb) multidrug-resistant(MDR) isolates, extensively drug-resistant(XDR) isolates, and the H37 RV strain. BP/CL activity against the H37 Rv strain was assessed in liquid cultures, in macrophages, and in mice. Results BP/CL exhibited activity against MDR and XDR Mtb isolates in liquid cultures. BP/CL treatment significantly reduced the number of colony forming units(CFU) of Mtb within macrophages compared with control untreated infected macrophages. Notably, BP/CL synergized in pairwise combinations with protionamide, aminosalicylate, and capreomycin to achieve a fractional inhibitory concentration for each pairing of 0.375 in vitro. In a mouse tuberculosis infection model, the efficacy of a cocktail of levofloxacin + pyrazinamide + protionamide + aminosalicylate against Mtb increased when the cocktail was combined with BP/CL, achieving efficacy similar to that of the positive control treatment(isoniazid + rifampin + pyrazinamide) after 2 months of treatment. Conclusion BP/CL may provide a new option to clinically treat MDR tuberculosis. 展开更多

Peptide bond synthesis is favorable to the production of bioactive small peptides.However,the abuse of toxic reagents remains an issue for chemical synthesis method,whereas the low product yield and purity limit the widespread use of enzymatic method.In this study,a new solid-phase enzymatic peptide synthesis(SPEPS) strategy was developed to produce an antioxidant tyrosine-alanine dipeptide(Tyr-Ala) by using recombinant carboxypeptidase Y(CPY) as the catalyst.The general SPEPS procedure involves three steps.First,the N-protected acyl donor was covalently attached to solid resin.Second,the peptide bond was condensed between the acyl donor and the nucleophile under the catalysis of CPY.Finally,one-step cleavage was performed to remove the protecting group and cleave the peptides from solid resin.Upon the optimization of reaction conditions,77.92%(±2.723%) yield of Tyr-Ala with high product purity of 90.971%(±2.695%) was obtained.In addition,the antioxidant activity of Tyr-Ala was determined by ABTS method,indicating that the synthesized Tyr-Ala obtained by SPEPS showed a superior antioxidant capability compared with commercial glutathione. 展开更多

目的探讨D-二聚体(D-dimer)在抗中性粒细胞胞浆抗体相关性血管炎(anti-neutrophil cytoplasmic antibody-associated vasculitis,AAV)患者中的临床意义。方法回顾性分析2014年5月至2017年11月我院肾内科穿刺活检确诊的50例ANCA相关性血... 目的探讨D-二聚体(D-dimer)在抗中性粒细胞胞浆抗体相关性血管炎(anti-neutrophil cytoplasmic antibody-associated vasculitis,AAV)患者中的临床意义。方法回顾性分析2014年5月至2017年11月我院肾内科穿刺活检确诊的50例ANCA相关性血管炎患者的临床资料。采用免疫比浊法检测血浆D-二聚体水平,以其中位数(3.17 mg/L FEU)为截点值将AAV患者分成高低水平2组,低水平组D-dimer<3.17 mg/L FEU,高水平组D-dimer≥3.17 mg/L FEU,比较2组患者临床特征、实验室检查、肾脏病理及预后差异。结果①高水平组患者C反应蛋白(C-reaction protein,CRP)、白细胞介素6(Interleukin-6,IL-6)水平及伯明翰血管炎活动性评分(Birmingham vasculitis activity score,BVAS)高于低水平组(P值分别为0.001、0.015、0.055),24小时尿蛋白定量、血清白蛋白(albumin,Alb)低于低水平组(P值分别为0.031、0.006)。②AAV患者血浆D-二聚体水平与BVAS(r=0.322,P=0.022)、CRP(r=0.573,P<0.001)、IL-6(r=0.341,P=0.015)、肿瘤坏死因子α(tumor necrosis factor alpha,TNF-α)(r=0.343,P=0.015)呈正相关。③肾脏病理特征:相较于高水平组患者,低水平组中球性硬化百分比较高(P=0.002),新月体形成的患者较少(P=0.247)。高水平组中细胞性新月体形成的患者数较低水平组多(P=0.017),且百分比高于低水平组(P=0.027)。④血浆D-二聚体(AUC=0.707)评估活动性AAV的预后无统计学差异(P>0.05)。结论血浆D-二聚体水平与AAV患者疾病活动性密切相关,有望成为评估AAV活动性的生物学标志物。 展开更多

A novel series of diaryl biuret derivatives containing a tetrazole moiety was designed and synthesized.All the target compounds were evaluated for their in vitro antitumor activity against HT-29,HepG2,MCF-7 and A549 cells by MTT assay.Most of them exhibited obvious antitumor activity,and four of them(4a,4c,4h and 7a)were superior to sorafenib in general.Among them,Compound 4h displayed more potent activity than sorafenib in all tested cancer cells.Compound 4c exhibited the most outstanding activity in inhibition of growth of HepG2 cells(IC50=0.55 μmol/L).Further,they both revealed favorable metabolic stability in in vitro assay.Compounds 4c and 4h are promising candidates for further development. 展开更多

This research was undertaken to clarify the characteristics of vegetation change and its main influencing factors on the Qinghai-Tibet Plateau. Using the greenness rate of change (GRC) and correlation factors, we analyzed the trend of vegetation change and its dominant factors from 2000 to 2015. The results indicate that the vegetation tended to improve from 2000 to 2015 on the Qinghai-Tibet Plateau, with the improved area accounting for 39.93% of the total;and the degraded area accounting for 19.32%. The areas of degraded vegetation are mainly concentrated in the low-relief and intermediate- relief mountains of the high-altitude and extremely high-altitude areas on the Qinghai-Tibet Plateau, as the vegetation characteristics are impacted by the terrain. Temperature and precipitation have obvious response mechanisms to vegetation growth, but the effects of precipitation and temperature on vegetation degradation are not significant over a short time frame. Overgrazing and population growth are the dominant factors of vegetation degradation on the Qinghai-Tibet Plateau. 展开更多

黔产薏苡仁油成分分析及对破骨细胞活性的影响 预览

目的:分析黔产薏苡仁油成分并研究不同浓度黔产薏苡仁油对破骨细胞活性的影响。方法:采用无水乙醇超声辅助法提取黔产薏苡仁油,并对其进行脂肪酸残基的气相色谱-质谱联用(GC-MS)分析,以雌二醇(10-8mol/L)作为阳性对照,采用原代分离新西... 目的:分析黔产薏苡仁油成分并研究不同浓度黔产薏苡仁油对破骨细胞活性的影响。方法:采用无水乙醇超声辅助法提取黔产薏苡仁油,并对其进行脂肪酸残基的气相色谱-质谱联用(GC-MS)分析,以雌二醇(10-8mol/L)作为阳性对照,采用原代分离新西兰乳兔骨髓细胞,并用1α,25-(OH)2Vit D3诱导获得破骨细胞,通过抗酒石酸酸性磷酸酶(TRAP)+细胞数、TRAP酶活及骨陷窝面积,分析黔产薏苡仁油对破骨细胞活性的影响。结果:通过GC-MS分析黔产薏苡仁油的脂肪酸残基主要为十四烷酸(肉豆蔻酸)、十六烷酸(棕榈酸)、十八烷酸(硬脂酸)、十八碳一烯酸(油酸)、十八碳二烯酸(亚油酸)、二十二烷酸(山嵛酸)、角鲨烯和γ-谷甾醇;进一步实验表明,黔产薏苡仁油各浓度组(0.98、1.95、3.91、7.81、15.63、31.25μg/mL)对破骨细胞活性具有一定的抑制作用,其中以高浓度组31.25μg/mL对破骨细胞的活性的抑制作用最为显著(p <0.05),且呈现出较为良好的浓度依赖性。结论:黔产薏苡仁油对破骨细胞活性具有抑制作用。 展开更多

了解一个城市群内各县(市)区的活跃度可以为该城市群服务设施的配置及等级结构的优化提供依据。基于环长株潭城市群夜间灯光数据、客流量数据以及新浪微博签到数据,利用数理统计分析,对环长株潭城市群内64个县(市)区的活跃度进行测度,... 了解一个城市群内各县(市)区的活跃度可以为该城市群服务设施的配置及等级结构的优化提供依据。基于环长株潭城市群夜间灯光数据、客流量数据以及新浪微博签到数据,利用数理统计分析,对环长株潭城市群内64个县(市)区的活跃度进行测度,并对经济活跃度和人口活跃度的组合状况进行分类分析。结果显示:长沙市内五区的活跃度居前五位,而距离各中心城区较远的县市区活跃度较低,总体上呈中心-外围的圈层分布,且具有沿轴向扩展与多极点并存的空间分布格局;核心区的经济活跃度与人口活跃度基本一致,而外围区则不一致;从经济活跃度和人口活跃度的组合关系来看,表现出高-高、高-低、低-高及低-低等4种类型。 展开更多

Jatrogricaine A(1), a new diterpenoid possessing a 5/6/6/4 carbon ring system, together with eight known diterpenoids(2-9) were isolated from the stems of Jatropha podagrica. Their structures were elucidated by extensive spectroscopic methods and the absolute configuration of 1 was determined by single crystal X-ray diffraction analysis. All compounds were evaluated for their anti-inflammatory activities in vitro, and compound 3 showed significant inhibitory effects against nitric oxide(NO) production in lipopolysaccharide(LPS)-induced RAW264.7 macrophage cells with an IC50 of 13.44±0.28 mmol×L^–1, being comparable to the positive control, quercetin(IC50 17.00±2.10 mmol×L^–1). 展开更多

地铁盾构渣土是城市轨道交通建设过程中大量产生的泥态废弃物。由于盾构渣土来源的广泛性、区域性、成分的复杂性,以及施工工艺的原因,使得盾构渣土回收利用困难,阻碍施工进度,污染城市环境。为进一步促进盾构渣土资源化再利用,本文主... 地铁盾构渣土是城市轨道交通建设过程中大量产生的泥态废弃物。由于盾构渣土来源的广泛性、区域性、成分的复杂性,以及施工工艺的原因,使得盾构渣土回收利用困难,阻碍施工进度,污染城市环境。为进一步促进盾构渣土资源化再利用,本文主要对我国盾构渣土资源化再利用途径的现状进行了分析;以郑州市地铁四号线、十四号线的渣土为例,测定了渣土的含水率及颗粒级配,并进行了XRD分析和差热分析,研究其矿物成分和物理特性随温度变化的情况。通过渣土水泥试块抗压强度与水泥净浆试块抗压强度的比值来反映渣土活性的变化,探讨了温度以及CaO、CaSO4·2H2O两种激发剂掺量对渣土活性的影响。研究表明:随着温度的升高,渣土水泥试块的抗压强度先减小后增大,在800℃时达到水泥净浆试块抗压强度的85.7%,满足Ⅱ级粉煤灰的抗压强度比要求。 展开更多

切开复位术对踝骨关节骨折合并脱位临床分析 预览

目的探讨踝骨关节骨折合并脱位的患者应用切开复位术方案治疗的效果及预后。方法选择踝骨关节骨折合并脱位的患者100例,均为我院骨科2017年5月—2018年5月收治,随机分组,对采用整复石膏外固定方案治疗(对照组,n=50)与采用切开复位内固... 目的探讨踝骨关节骨折合并脱位的患者应用切开复位术方案治疗的效果及预后。方法选择踝骨关节骨折合并脱位的患者100例,均为我院骨科2017年5月—2018年5月收治,随机分组,对采用整复石膏外固定方案治疗(对照组,n=50)与采用切开复位内固定术方案治疗(观察组,n=50)效果及预后展开对比。结果观察组手术总有效率为94%,高于对照组的72%,差异具有统计学意义(P<0.05);观察组住院天数、骨折愈合天数、负重天数均少于对照组,差异具有统计学意义(P<0.05);观察组治疗后踝关节功能评分高于对照组,关节活动度恢复优于对照组,差异均具有统计学意义(P<0.05)。结论针对临床收治的踝骨关节骨折合并脱位的患者,应用切开复位术予以治疗,可显著增强手术效果,缩短康复进程,改善踝关节功能,具有非常重要的实施价值。 展开更多
