Clinical and muscle magnetic resonance image findings in patients with late-onset multiple acyl-CoA dehydrogenase deficiency

Background:Late-onset multiple acyl-coA dehydrogenase deficiency (MADD) is an autosomal recessive inherited metabolic disorder. It is still unclear about the muscle magnetic resonance image (MRI) pattern of the distal lower limb pre- and post-treatment in patients with late-onset MADD. This study described the clinical and genetic findings in a cohort of patients with late-onset MADD, and aimed to characterize the MRI pattern of the lower limbs.Methods:Clinical data were retrospectively collected from clinic centers of Peking University People's Hospital between February 2014 and February 2018. Muscle biopsy, blood acylcarnitines, and urine organic acids profiles, and genetic analysis were conducted to establish the diagnosis of MADD in 25 patients. Muscle MRI of the thigh and leg were performed in all patients before treatment. Eight patients received MRI re-examinations after treatment.Results:All patients presented with muscle weakness or exercise intolerance associated with variants in the electron transfer flavoprotein dehydrogenase gene. Muscle MRI showed a sign of both edema-like change and fat infiltration selectively involving in the soleus (SO) but sparing of the gastrocnemius (GA) in the leg. Similar sign of selective involvement of the biceps femoris longus (BFL) but sparing of the semitendinosus (ST) was observed in the thigh. The sensitivity and specificity of the combination of either "SO+/GA-" sign or "BFL+/ST-" sign for the diagnosis of late-onset MADD were 80.0% and 83.5%, respectively. Logistic regression model supported the findings. The edema-like change in the SO and BFL muscles were quickly recovered at 1 month after treatment, and the clinical symptom was also relieved.Conclusions:This study expands the clinical and genetic spectrums of late-onset MADD. Muscle MRI shows a distinct pattern in the lower limb of patients with late-onset MADD. The dynamic change of edema-like change in the affected muscles might be a potential biomarker of treatment response. 展开更多

BACKGROUND Thrombocytopenia associated with acute kidney injury is a challenging disorder. Thrombotic microangiopathy (TMA) is a potentially life- or organ-threatening syndrome that can be induced by several disorders or medical interventions. There is overlap between the clinical presentation and pathophysiology of thrombotic thrombocytopenia purpura and hemolytic uremic syndrome (HUS), and to a lesser extent, disseminated intravascular coagulation (DIC). We describe a case to illustrate the potential diagnostic difficulty, especially at initial presentation. CASE SUMMARY We reported a case of a 44-year-old woman that presented with diarrhea, thrombocytopenia, schistocytes, elevated serum lactate dehydrogenase (LDH) level and acute kidney injury. While the clinical presentation resembled that of Shiga toxin–induced HUS, the disease course was more consistent with gastrointestinal infection-related DIC. To aid in the accurate diagnosis of TMA and other associated disorders, we have undertaken a review and provided a clear interpretation of some typical biomarkers including schistocytes, LDH and platelet count, coagulation profile and more specific indexes of ADAMTS13, complement profile, and the isolation of Shiga toxin-producing Escherichia coli (commonly referred to as STEC). CONCLUSION The use and correct interpretation of classical indexes of schistocyte, LDH, and platelet count is vital in diagnosing TMA and associated disorders. Understanding the characteristics of these biomarkers in the context of thrombocytopenia purpura, HUS and DIC will facilitate the accurate diagnosis and early initiation of appropriate treatment. 展开更多

MBR技术中剩余污泥减量机理研究 预览

由于剩余污泥量大,已成为制约污水生物处理技术发展的重要因素.膜生物反应器(MBR)作为新型高效污水处理技术,能够实现污水处理中的剩余污泥减量.采用对比法,对MBR技术中剩余污泥减量的机理进行了研究.研究表明,溶解氧(DO)质量浓度在1.0~... 由于剩余污泥量大,已成为制约污水生物处理技术发展的重要因素.膜生物反应器(MBR)作为新型高效污水处理技术,能够实现污水处理中的剩余污泥减量.采用对比法,对MBR技术中剩余污泥减量的机理进行了研究.研究表明,溶解氧(DO)质量浓度在1.0~3.0 mg/L内,污染物处理效果均可达到90%以上,污泥浓度(MLVSS)可控制在10 g/L左右,脱氢酶活性平均值为136.82 mg/g-VSS,膜区污泥中三磷酸腺苷(ATP)含量平均为0.003 56μg/mg-VSS.分析发现,在MBR中发生了能量解偶联作用,膜区附近、反应器中部、进水口附近的胞外聚合物(EPS)含量分别为15.15、12.79、15.38 mg/g-VSS,EPS和胞外蛋白酶相互作用,EPS、微生物残骸等大分子物质被高活性的胞外蛋白酶降解,成为可以被微生物代谢的基质,促进了微生物隐性增长,从而有效降低了污泥产率,实现了剩余有机污泥减量. 展开更多

不同测定方法探讨重金属对微生物毒性的影响 预览

微生物是污水处理工艺中污染物的重要分解者,重金属对活性污泥微生物具有一定毒性。为了研究和比较重金属对活性污泥微生物的毒性影响,采用发光细菌毒性、活性污泥脱氢酶毒性、硝化抑制毒性3种方法测定4种重金属(Hg、Cd、Zn、Pb)对活性... 微生物是污水处理工艺中污染物的重要分解者,重金属对活性污泥微生物具有一定毒性。为了研究和比较重金属对活性污泥微生物的毒性影响,采用发光细菌毒性、活性污泥脱氢酶毒性、硝化抑制毒性3种方法测定4种重金属(Hg、Cd、Zn、Pb)对活性污泥微生物的毒性,并对测定结果进行了比较。结果表明,4种重金属均可降低污泥系统CODCr的去除率,其中,Cd和Hg对CODCr去除率的影响最明显(43.2%-92.3%和52.4%-92.5%),而Pb和Zn对活性污泥CODCr去除率的影响较小。发光细菌毒性测定方法的灵敏度最高(达50%以上),测得的重金属半数有效浓度(EC50)最低(Hg9.6mg·L^-1,Cd0.3mg·L^-1,Zn3.6mg·L^-1,Pb13.9mg·L^-1),4种重金属对发光细菌发光强度的抑制程度表现为Hg >Cd >Zn >Pb,发光细菌的发光强度抑制率与重金属的对数浓度呈极显著的线性相关关系(P<0.01),发光细菌发光强度随重金属对数浓度的增大而减小。活性污泥脱氢酶毒性和硝化抑制毒性的测定结果与发光细菌毒性测定结果相比,灵敏度相对较低,测得的重金属EC50相对较高,活性污泥脱氢酶活性的抑制程度大小顺序为Cd >Hg >Zn >Pb,与测得的活性污泥硝化速率抑制程度大小顺序一致,但二者测得的EC50有所差别。4种重金属与活性污泥脱氢酶活性的Logistic回归方程均达到极显著水平(P<0.01),活性污泥SVI值随重金属浓度的增加而升高,当重金属浓度较低时,其对活性污泥SVI的影响并不大。为了更准确地判定重金属对活性污泥微生物的毒性影响,应取不同的重金属毒性终点指示指标进行毒性组实验,而不能以发光细菌毒性的单一测定结果作为评判依据 展开更多

The current method for DHA production is Gluconobacter oxydans ( G. oxydans ) fermentation, but the high concentration of glycerol in the fermentation broth inhibits cells growth. To overcome this obstacle, in this study, we overexpressed the glycerol transporter (GlpFp) by the use of promoters P tufB , P gmr , P glp1 , and P glp2 in G. oxydans 621H. The results show that the glycerol tolerances of strains overexpressing G lpF were all much better than that of the control strain. The glycerol dehydrogenase gene (G dh) was overexpressed by the promoters P tufB and P gdh , which increased the DHA titer by 12.7% compared with that of the control group. When G lpF and Gdh genes were co-overexpressed in G. oxydans 621H, the OD600 value of the engineered strains all increased, but the DHA titers decreased in di erent degrees, as compared with strains that overexpressed only G dh . Rdh13 deficiency weakens carbon tetrachloride-induced liver injury by regulating Spotl4 and Cyp2e1 expression levels

Mitochondrion-localized retinol dehydrogenase 13 (Rdh13) is a short-chain dehydrogenase/reductase involved in vitamin A metabolism in both humans and mice. We previously generated Rdh13 knockout mice and showed that Rdh13 deficiency causes severe acute retinal light damage. In this study, considering that Rdh13 is highly expressed in mouse liver, we further evaluated the potential effect of Rdh13 on liver injury induced by carbon tetrachloride (CC14). Although Rdh13 deficiency showed no significant effect on liver histology and physiological functions under regular culture, the Rdh13^-/- mice displayed an attenuated response to CCl4-induced liver injury. Their livers also exhibited less histological changes and contained lower levels of liver-related metabolism enzymes compared with the livers of wild-type (WT) mice. Furthermore, the Rdhl3 1 mice had Rdh13 deficiency and thus their liver cells were protected from apoptosis, and the quantity of their proliferative cells became lower than that in WT after CC14 exposure. The ablation of Rdhl3 gene decreased the expression levels of thyroid hormone-inducible nuclear protein 14 (Spot14) and cytochrome P450 (Cyp2el) in the liver, especially after CC14 treatment for 48 h. These data suggested that the alleviated liver damage induced by CC14 in Rdh13^-/- mice was caused by Cyp2el enzymes, which promoted reductive CC14 metabolism by altering the status of thyroxine metabolism. This result further implicated Rdhl3 as a potential drug target in preventing chemically induced liver injury. 展开更多

White adipose tissue (WAT) stores energy and also plays an important endocrine role in producing adipokines for communication with the peripheral and central nervous system. WAT consists of the major lipogenic unilocular adipocytes and the minor populations of beige and brite multilocular adipocytes. These multilocular adipocytes express thermogenic genes and have phenotypic similarity with thermogenic brown adipose tissue. According to a current paradigm, multilocular adipocytes have a thermogenic function in WAT. In this mini review, we discuss data revealing heterogeneity among multilocular cell subsets in WAT and their functions beyond thermogenesis. We propose a hypothetical neuroendocrine role for multilocular adipocytes subsets in the formation of adaptive sensory-sympathetic circuits between the central nervous system and adipose tissue, which activate lipolysis and thermogenesis in WAT in high energy demand situations. 展开更多

Xanthine dehydrogenase(XDH) is a crucial enzyme involved in purine metabolism. To evaluate the effect of XDH deficiency on rice growth during dark treatment, wild type(WT) Nipponbare(Oryza sativa L.) and two independent transgenic lines with severe RNAi suppression(xdh3 and xdh4) were used in the present experiment. Under normal growth conditions, chlorophyll levels and biomass were indistinguishable between WT and the two RNAi transgenic lines, but XDH enzyme activity and ureide levels were suppressed in XDH RNAi transgenic lines. When XDH RNAi transgenic lines were subjected to dark treatment, chlorophyll content and biomass were significantly decreased, while O2–· production rate and malonaldehyde(MDA) were significantly increased compared to WT. The spraying test of exogenous allantoin raised chlorophyll content and biomass and reduced O2–· production rate and MDA in WT and both transgenic lines, and it also simultaneously reduced differences between RNAi and WT plants caused by XDH deficiency in growth potential and anti-oxidative capacity under dark treatment. These results suggested that fully functional purine metabolism plays an important role in reducing the sensitivity of rice seedlings to dark stress. 展开更多

随着磺胺类抗生素(Sulfonamides)的大规模生产和使用,其对环境的危害日益加重。通过试验室盆栽试验的方法,研究了3种典型磺胺类抗生素磺胺甲恶唑(SMZ)、磺胺甲基嘧啶(SM1)、磺胺噻唑(ST),在不同染毒剂量下,单一与两两复合污染对油菜种... 随着磺胺类抗生素(Sulfonamides)的大规模生产和使用,其对环境的危害日益加重。通过试验室盆栽试验的方法,研究了3种典型磺胺类抗生素磺胺甲恶唑(SMZ)、磺胺甲基嘧啶(SM1)、磺胺噻唑(ST),在不同染毒剂量下,单一与两两复合污染对油菜种植土壤脱氢酶(DHA)和过氧化氢酶(CAT)活性的影响。结果表明,SMZ和SM1均对DHA活性产生抑制作用,而ST对DHA的活性呈现诱导作用,并且随着染毒质量比的增加,作用效应均呈增强趋势。SM1与ST对CAT活性产生诱导作用,同时随染毒质量比增加,诱导作用明显减弱。而SMZ对CAT的活性呈抑制作用,低质量比时,这种抑制作用不显著(p<0.05),高质量比时,SMZ对CAT的活性产生极显著(p<0.01)抑制作用。复合组并未显著增强对DHA和CAT的毒性效应,复合污染对土壤酶活性的效应与染毒质量比及土壤酶的种类等均相关。由以上可见,用土壤酶活性指标来表征土壤磺胺类抗生素污染情况是可行的。 展开更多

Aldehyde dehydrogenase 2(ALDH2)is best known for its critical detoxifying role in liver alcohol metabolism.However,ALDH2 dysfunction is also involved in a wide range of human pathophysiological situations and is associated with complications such as cardiovascular diseases,diabetes mellitus,neurodegenerative diseases and aging.A growing body of research has shown that ALDH2 provides important protection against oxidative stress and the subsequent loading of toxic aldehydes such as 4-hydroxy-2-nonenal and adducts that occur in human diseases,including ischemia reperfusion injury(IRI).There is increasing evidence of its role in IRI pathophysiology in organs such as heart,brain,small intestine and kidney;however,surprisingly few studies have been carried out in the liver,where ALDH2 is found in abundance.This study reviews the role of ALDH2 in modulating the pathways involved in the pathophysiology of IRI associated with oxidative stress,autophagy and apoptosis.Special emphasis is placed on the role of ALDH2 in different organs,on therapeutic“preconditioning”strategies,and on the use of ALDH2 agonists such as Alda-1,which may become a useful therapeutic tool for preventing the deleterious effects of IRI in organ transplantation. 展开更多

Poikilothermic animals living in high-altitude environments can be greatly affected by the anaerobic metabolism and lactate recycling,which are catalyzed by an enzyme called lactate dehydrogenase (LDH).However, the function and possible regulatory mechanisms of their anaerobic glycolysis remained elusive.We compared the difference in LDH between a native high-altitude (4353m)lizard,Phrynocephalus erythrurus,and a closely related species,Phrynocephalusprzewalskii that lives in intermediate altitude environment (1400m).The activity of LDH,the concentration of lactate,the distribution of isoenzyme,and the mRNA amounts of Ldh-A and Ldh-B were determined. In cardiac muscle,the lactate-forming activity of P.erythrurus in LDH was higher than of P.przewalskii LDH at all three temperatures tested (10℃,25℃ and 35℃),while lactate-oxidation activity of LDH was significantly different between the two species only at 25℃ and 35℃.In skeletal muscle,both lactate-forming and lactate-oxidation rates of P.erythrurus were lower than that of P.przewalskii.There was a higher proportion ofH subunit and a significantly higher expression of Ldh-B,with a concomitant decrease of lactate concentration in P.erythrurus.These results indicate that P. erythrurus may have a strong potential for anaerobic metabolism,which is likely adapted to the hypoxic environment at high altitudes.Furthermore,P.erythrurus is capable of oxidizing more lactate than P.przewalskii.The Ldh-A cDNA of the two species consists of a 999bp open reading frame (ORF),which encodes 332 amino acids,while Ldh-B cDNA consists of a 1002 bp ORF encoding 333amino acids.LDHA has the same amino acid sequence between the two species, but three amino acid substitutions (V12I,N21S and N318K)were observed in LDHB.Structure analysis of LDH indicated that the substitutions of residues Val12 and Asp21 in P.erythrurus could be responsible for the highaltitude adaptation.The LDH characteristics of LDH in P.erythrurus suggest unique adaptation strategies of anaerobic metabolism in 展开更多

Indication of Soil Microbial Activity by Electrical Signals of Microbial Fuel Cells with Re-Vegetated Red Soils

Electrical signals generated by microbial fuel cells (MFCs) may potentially indicate overall soil microbial activity. This study aimedto validate whether a significant correlation existed between soil microbial activity and electrical signals in different soils. Re-vegetatedred soils with four vegetation types and unplanted eroded red soil were collected and packed into MFCs. The voltage of the MFCs wasrecorded during operation. Electrical signals including start-up time (ST), peak voltage, and coulomb quantity were extracted fromthe voltage curves. Soil dehydrogenase activity (DHA) was determined as a reference method to verify the reliability of the electricalsignals. After MFC operation, the anodic bacterial community was analyzed by Illumina sequencing based on the 16S rRNA gene. Theresults indicated that ST was significantly correlated with DHA, along with soil organic carbon content. The linear regression modeldescribed the optimal relationships for ST and DHA. Clostridium and Acinetobacter constituted the dominant genera on the anodeof the MFCs. The results demonstrated that ST may indicate soil microbial activitv in re-vegetated red soils. 展开更多
