Kai Xin San(KXS,containing ginseng,hoelen,polygala,and acorus),a traditional Chinese herbal compound,has been found to regulate cognitive dysfunction;however,its mechanism of action is still unclear.In this study,72 specific-pathogen-free male Kunming mice aged 8 weeks were randomly divided into a vehicle control group,scopolamine group,low-dose KXS group,moderate-dose KXS group,high-dose KXS group,and positive control group.Except for the vehicle control group and scopolamine groups(which received physiological saline),the doses of KXS(0.7,1.4 and 2.8 g/kg per day)and donepezil(3 mg/kg per day)were gastrointestinally administered once daily for 2 weeks.On day 8 after intragastric treatment,the behavioral tests were carried out.Scopolamine group and intervention groups received scopolamine 3 mg/kg per day through intraperitoneal injection.The effects of KXS on spatial learning and memory,pathological changes of brain tissue,expression of apoptosis factors,oxidative stress injury factors,synapse-associated protein,and cholinergic neurotransmitter were measured.The results confirmed the following.(1)KXS shortened the escape latency and increased residence time in the target quadrant and the number of platform crossings in the Morris water maze.(2)KXS increased the percentage of alternations between the labyrinth arms in the mice of KXS groups in the Y-maze.(3)Nissl and terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling staining revealed that KXS promoted the production of Nissl bodies and inhibited the formation of apoptotic bodies.(4)Western blot assay showed that KXS up-regulated the expression of anti-apoptotic protein Bcl-2 and inhibited the expression of pro-apoptotic protein Bax.KXS up-regulated the expression of postsynaptic density 95,synaptophysin,and brain-derived neurotrophic factor in the cerebral cortex and hippocampus.(5)KXS increased the level and activity of choline acetyltransferase,acetylcholine,superoxide dismutase,and glutathione peroxidase,and reduced the level and activity o 展开更多

G protein-coupled receptor kinase 2 inhibition improves erectile function through amelioration of endothelial dysfunction and oxidative stress in a rat model of type 2 diabetes

Type 2 diabetes mellitus (T2DM)is a common cause of erectile dysfunction (ED).It has been demonstrated that G protein-coupled receptor kinase 2 (GRK2)overexpression contributes to diabetic endothelial dysfunction and oxidative stress,which also underlies ED in T2DM.We hypothesized that GRK2 overexpressed and attenuated endothelial function of the cavernosal tissue in a rat model of T2DM.T2DM rats were established by feeding with a high-fat diet (HFD)for 2 weeks and then administering two intraperitoneal (IP) injections of a low dose of streptozotocin (STZ),followed by continuous feeding with a HFD for 6 weeks.GRK2 was inhibited by IP injection of paroxetine,a selective GRK2 inhibitor,after STZ injection.Insulin challenge tests,intracavernous pressure (ICP), GRK2 expression,the protein kinase B (Akt)/endothelial nitric oxide synthase (eNOS)pathway,nicotinamide adenine dinucleotide phosphate (NADPH)oxidase subunit gp91phox,nitric oxide (NO),reactive oxygen species (ROS)production,and apoptosis in cavernosal tissue were examined.Less response to insulin injection was observed in T2DM rats 2 weeks after HFD.Markedly increased GRK2 expression,along with impaired Akt/eNOS pathway,reduced NO production,increased gp91phox expression and ROS generation,increased apoptosis and impaired erectile function were found in T2DM rats.inhibition of GRK2 with paroxetine ameliorated Akt/eNOS signaling,restored NO production,downregulated NADPH oxidase,subsequently inhibited ROS generation and apoptosis,and ultimately preserved erectile function.These results indicated that GRK2 upregulation may be an important mechanism underlying T2DM ED,and GRK2 inhibition may be a potential therapeutic strategy for T2DM ED. 展开更多

Sexual dysfunction is a disruption of normal sexual intercourse and may be a consequence of various organic and psychological disorders. It is the most frequent at patients with cardiovascular and neurological diseases, sedentary lifestyle, arterial hypertension, diabetes, obesity and metabolic syndrome, and may disrupt their emotional life. But, sexual dysfunction may occur simply because of the physical aging of the body. Physiologic changes that occur with aging will aff ect sexual function and could be exacerbated by the above-mentioned diseases. Except for the sexual counseling and standard medications (phosphodiesterase type 5 (PDE5)– inhibitors, local estrogen therapy), some of the herbal medicines (aphrodisiacs) may be helpful in the treatment of these patients making them emotionally younger and happier. In this review, we bring a list of the pharmacologically tested aphrodisiac plants. But, we have a lack of information about their mechanisms of action, drug interaction and safety profi le. Until the relevant information is collected, uses of these products may be risky to the human being and should be prescribed only in consultation with a specialist. 展开更多

BACKGROUND Early allograft dysfunction(EAD)after liver transplantation(LT)is an important cause of morbidity and mortality.To ensure adequate graft function,a critical hepatocellular mass is required in addition to an appropriate blood supply.We hypothesized that intraoperative measurement of portal venous and hepatic arterial flow may serve as a predictor in the diagnosis of EAD.AIM To study whether hepatic flow is an independent predictor of EAD following LT.METHODS This is an observational cohort study in a single institution.Hepatic arterial blood flow and portal venous blood flow were measured intraoperatively by transit flow.EAD was defined using the Olthoff criteria.Univariate and multivariate analyses were used to determine the intraoperative predictors of EAD.Survival analysis and prognostic factor analysis were performed using the Kaplan-Meier and Cox regression models.RESULTS A total of 195 liver transplant procedures were performed between January 2008 and December 2014 in 188 patients.A total of 54(27.7%)patients developed EAD.The median follow-up was 39 mo.Portal venous flow,hepatic arterial flow(HAF)and total hepatic arterial flow were associated with EAD in both the univariate and multivariate analyses.HAF is an independent prognostic factor for 30-d patient mortality.CONCLUSION Intraoperative measurement of blood flow after reperfusion appears to be a predictor of EAD;Moreover,HAF should be considered a predictor of 30-d patient mortality. 展开更多

OBJECTIVE: To investigate neuropsychological features of post-stroke cognitive impairment with no dementia(PSCIND) patients with different Traditional Chinese Medicine(TCM) syndromes.METHODS: We recruited 50 patients with PSCIND between April 2012 and March 2013. Patients were divided into different groups according to TCM classifications. Patients were assessed using neuropsychological tests, including cognitive screening(mini-mental state examination), memory testing (auditory verbal learning test), executive/attention[shape trails test, stroop color-word test(SCWT),reading the mind in the eyes test, the digit ordering test-A(DOT-A), and symbol digit modalities test], language(action naming test, Boston naming test, famous face test, similarity test, and verbal fluency test), and visuospatial functioning [complex figure test(CFT)].RESULTS: We found no significant differences between patients with and without a diagnosis of turbid phlegm blocking the upper orifices on neuropsychological test performance. Patients diagnosed with upper hyperactivity of liver Yang syndrome scored significantly lower on the SCWT-C executive test and the CFT-delayed recall memory test. Patients with excess syndrome scored significantly lower on the SCWT-C executive test, and significantly higher on the DOT-A executive test.CONCLUSION: Neuropsychological characteristics differ between PSCIND patients with different TCM classifications. 展开更多

目的 探讨护理干预模式对手外伤患者术后疗效的影响。方法 80例行手外伤手术患者,按照随机数字表法分为对照组和观察组,各40例。对照组接受常规护理,观察组在常规护理基础上加强护理干预,比较两组患者的护理效果。结果 干预前,两组患者... 目的 探讨护理干预模式对手外伤患者术后疗效的影响。方法 80例行手外伤手术患者,按照随机数字表法分为对照组和观察组,各40例。对照组接受常规护理,观察组在常规护理基础上加强护理干预,比较两组患者的护理效果。结果 干预前,两组患者视觉模拟评分法(VAS)评分比较,差异无统计学意义(P>0.05);干预后,观察组患者VAS评分低于对照组,差异有统计学意义(P<0.05)。观察组患者术后手功能恢复优良率为92.5%,明显高于对照组的77.5%,差异有统计学意义(P<0.05)。结论 针对手外伤手术患者应用护理干预,可以有效缓解患者术后的疼痛程度,提高手功能的恢复效果,预防手功能出现障碍,值得推广和应用。 展开更多

电针配合运动疗法治疗尺神经损伤修复术后功能障碍

目的:观察电针配合运动疗法对尺神经损伤修复术后功能障碍的影响。方法:采用简单随机抽签法将60例尺神经损伤修复术后患者分为对照组(n=30)和观察组(n=30),观察组术后予以电针配合运动疗法,对照组术后仅予以运动疗法;比较两组术后12周... 目的:观察电针配合运动疗法对尺神经损伤修复术后功能障碍的影响。方法:采用简单随机抽签法将60例尺神经损伤修复术后患者分为对照组(n=30)和观察组(n=30),观察组术后予以电针配合运动疗法,对照组术后仅予以运动疗法;比较两组术后12周腕关节活动度、患手握力、患手/健手握力百分比、肩臂手残疾量表(DASH)值、明尼苏达手灵巧度(MMDT)评定值。结果:术后12周时,观察组腕关节掌区、背伸、尺偏、桡偏等状态的活动度及患手握力、患手/健手握力百分比均明显大于对照组,DASH值、MMDT评定值均明显小于对照组,差异有统计学意义(P<0.05)。结论:电针配合运动疗法能够促进尺神经损伤修复术后患者手部功能恢复,是疗效确切的康复治疗方案。 展开更多

Hydrogen sulfide is an antioxidant molecule that has a wide range of biological effects against oxidative stress. Balanced oxidative stress is also vital for maintaining cellular function in biological system, where reactive oxygen species are the main source of oxidative stress. When the normal redox balance is disturbed, deoxyribonucleic acid, lipid, and protein molecules are oxidized under pathological conditions, like diabetes mellitus that leads to diabetic peripheral neuropathy. In diabetes mellitus-induced diabetic peripheral neuropathy, due to hyperglycemia, pancreatic beta cell(β cell) shows resistance to insulin secretion. As a consequence, glucose metabolism is disturbed in neuronal cells which are distracted from providing proper cell signaling pathway. Not only diabetic peripheral neuropathy but also other central damages occur in brain neuropathy. Neurological studies regarding type 1 diabetes mellitus patients with Alzheimer’s disease, Parkinson’s disease, and amyotrophic lateral sclerosis have shown changes in the central nervous system because high blood glucose levels(HbA1 c) appeared with poor cognitive function. Oxidative stress plays a role in inhibiting insulin signaling that is necessary for brain function. Hydrogen sulfide exhibits antioxidant effects against oxidative stress, where cystathionine β synthase, cystathionine γ lyase, and 3-mercaptopyruvate sulfurtransferase are the endogenous sources of hydrogen sulfide. This review is to explore the pathogenesis of diabetes mellitus-induced diabetic peripheral neuropathy and other neurological comorbid disorders under the oxidative stress condition and the anti-oxidative effects of hydrogen sulfide. 展开更多

Potential of phloroglucinol to improve erectile dysfunction associated with streptozotocin-induced diabetes in rats

Often, people living with diabetes develop erectile dysfunction (ED). The primary aim of this work was to investigate the effect of phloroglucinol in diabetes-induced ED in rats. Methods: Male Wistar rats were given 52 mg/kg of streptozotocin, by intraperitoneal injection, to induce diabetes and ED. Subsequently, animals were grouped into three groups: group 1, diabetic control;group 2, low-dose phloroglucinol (150 mg/kg body weight);and group 3, high-dose phloroglucinol (250 mg/kg body weight). A group of six normal rats served as a normal control. The rats were treated with phloroglucinol for six weeks and then were assessed for treatment effects. Sexual behavior, glycosylated hemoglobin Ale (HbAlc) values, serum testosterone, serum nitric oxide (NO), blood pressure and sperm count were measured after the end of treatment. Results: After a 6-week treatment period, the high dose of phloroglucinol significantly decreased HbAlc values in diabetic rats. Rats treated with phloroglucinol had increased serum testosterone, NO and sperm count. Animals treated with 250 mg/kg phloroglucinol performed similar to normal rats in the sexual behavioral study, suggesting the reversal of complications of ED. Conversely, a decrease in the blood pressure in treated groups was observed. Conclusion: The results highlight the protective effect of phloroglucinol in diabetes-induced ED in rats warranting further studies. 展开更多

Theα5 subunit-containing gamma-amino butyric acid type A receptors(α5 GABAARs)are a distinct subpopulation that are specifically distributed in the mammalian hippocampus and also mediate tonic inhibitory currents in hippocampal neurons.These tonic currents can be enhanced by low-dose isoflurane,which is associated with learning and memory impairment.Inverse agonists ofα5 GABAARs,such as L-655,708,are able to reverse the short-term memory deficit caused by low-dose isoflurane in young animals.However,whether these negative allosteric modulators have the same effects on aged rats remains unclear.In the present study,we mainly investigated the effects of L-655,708 on low-dose(1.3%)isoflurane-induced learning and memory impairment in elderly rats.Young(3-month-old)and aged(24-month-old)Wistar rats were randomly assigned to receive L-655,708 0.5 hour before or 23.5 hours after 1.3%isoflurane anesthesia.The Morris Water Maze tests demonstrated that L-655,708 injected before or after anesthesia could reverse the memory deficit in young rats.But in aged rats,application of L-655,708 only before anesthesia showed similar effects.Reverse transcription-polymerase chain reaction showed that low-dose isoflurane decreased the mRNA expression ofα5 GABAARs in aging hippocampal neurons but increased that in young animals.These findings indicate that L-655,708 prevented but could not reverse 1.3%isoflurane-induced spatial learning and memory impairment in aged Wistar rats.All experimental procedures and protocols were approved by the Experimental Animal Ethics Committee of Academy of Military Medical Science of China(approval No.NBCDSER-IACUC-2015128)in December 2015. 展开更多

Prolonged activation of adenosine A1 receptor likely leads to damage of dopaminergic neurons and subsequent development of neurodegenerative diseases.However,the pathogenesis underlying long-term adenosine A1 receptor activation-induced neurodegeneration remains unclear.In this study,rats were intraperitoneally injected with 5 mg/kg of the adenosine A1 receptor agonist N6-cyclopentyladenosine(CPA)for five weeks.The mobility of rats was evaluated by forced swimming test,while their cognitive capabilities were evaluated by Y-maze test.Expression of sortilin,α-synuclein,p-JUN,and c-JUN proteins in the substantia nigra were detected by western blot analysis.In addition,immunofluorescence staining of sortilin andα-synuclein was performed to detect expression in the substantia nigra.The results showed that,compared with adenosine A1 receptor antagonist 8-cyclopentyl-1,3-dipropylxanthine(5 mg/kg)+CPA co-treated rats,motor and memory abilities were reduced,surface expression of sortin andα-synuclein in dopaminergic neurons was reduced,and total sortilin and totalα-synuclein were increased in CPA-treated rats.MN9D cells were incubated with 500 nM CPA alone or in combination with 10μM SP600125(JNK inhibitor)for 48 hours.Quantitative real-time polymerase chain reaction analysis of sortilin andα-synuclein mRNA levels in MN9D cells revealed upregulated sortilin expression in MN9D cells cultured with CPA alone,but the combination of CPA and SP600125 could inhibit this expression.Predictions made using Jasper,PROMO,and Alibaba online databases identified a highly conserved sequence in the sortilin promoter that was predicted to bind JUN in both humans and rodents.A luciferase reporter assay of sortilin promoter plasmid-transfected HEK293T cells confirmed this prediction.After sortilin expression was inhibited by sh-SORT1,expression of p-JUN and c-JUN was detected by western blot analysis.Long-term adenosine A1 receptor activation levels upregulatedα-synuclein expression at the post-transcriptional level by affecting sort 展开更多

We aimed to determine whether combination of LIM-kinase 2 inhibitor(LIMK2i)and phosphodiesterase type-5 inhibitor(PDE5i)could restore erectile function through suppressing cavernous fibrosis and improving caver nous apoptosis in a rat model of cavernous nerve crush injury(CNCI).Seventy 12-week-old Sprague-Dawley rats were equally distributed into five groups as follows:(1)sham surgery(Group S),(2)CNCI(Group I),(3)CNCI treated with daily intraperitoneal administration of 10.0 mg kg^-1 LIMK2i(Group I+L),(4)daily oral administration of 20.0 mg kg-1 udenafil,PDE5i(Group I+U),and(5)combined administration of 10.0 mg kg^-1 LIMK2i and 20.0 mg kg^-1 udenafil(Group I+L+U).Rats in Groups I+L,I+U,and I+L+U were treated with respective regimens for 2 weeks after CNCI.At 2 weeks after surgery,erectile response was assessed using electrostimulation.Penile tissues were processed for histological studies and western blot.Group I showed lower intracavernous pressure(ICP)/mean arterial pressure(MAP),lower area under the curve(AUC)/MAP,decreased immunohistochemical staining for alpha-smooth muscle(SM)actin,higher apoptotic index,lower SM/collagen ratio,increased phospho-LIMK2-positive fibroblasts,decreased protein kinase B/endothelial nitric oxide synthase(Akt/eNOS)phosphorylation,increased LIMK2/cofilin phosphorylation,and increased protein expression of fibronectin,compared to Group S.In all three treatment groups,erectile responses,protein expression of fibronectin,and SM/collagen ratio were improved.Group I+L+U showed greater improvement in erectile response than Group I+L.SM content and apoptotic index in Groups I+U and I+L+U were improved compared to those in Group I.However,Group I+L did not show a significant improvement in SM content or apoptotic index.The number of phospho-LIMK2-positive fibroblasts was normalized in Groups I+L and I+L+U,but not in Group I+U.Akt/eNOS phosphorylation was improved in Groups I+U and I+L+U,but not in Group I+L.LIMK2/cofilin phosphorylation was improved in Groups I+L and I+L+U,but not in Group 展开更多

Vacuum erecti on device(VED),used to treat radical prostatectomy(RP)-associated erectile dysfunction,has attracted con siderable attention.However,the optimal negative pressure remains to be determined.This investigation explored the optimal pressure for VED therapy in penile rehabilitation.Thirty-six 9-week-old male rats were randomly divided into six groups:control groups(sham group,bilateral cavernous nerve crush[BCNC]group)and VED therapy groups(-200 mmHg group,-300 mmHg group,-400 mmHg group,-500 mmHg group).BCNC group and VED therapy groups underwent BCNC surgery.Intracavernosal pressure(ICP)/mean arterial pressure(MAP)ratio was calculated to assess erectile function.Mass on's trichrome(MT)staining,terminal deoxynucleotidyl transferase dUTP nick end labeling(TUNEL)assay,immunohistochemistry,and real-time polymerase chain reaction(RT-PCR)were performed to explore cellular and molecular changes of the penis.Compared to the BCNC group,ICP/MAP ratios in all VED treatment groups were improved significantly(all P<0.05),but there were no statistically significant differences among VED therapy groups.With increased pressure,complications gradually emerged and increased in frequency.Expression of molecular indicators,such as endothelial nitric oxide synthase(eNOS)and alpha-smooth muscle actin(a・SMA),increased after VED therapy,and hypoxia-inducible factor la(HIF-la)and transforming growth factor beta(TGF-p)decreased.In addition,VED therapy improved the outcomes of MT and TUNEL assay.This investigation demonstrated a pressure of-200 mmHg in a rat model is optimal for VED therapy for penile rehabilitation after RP.No further benefits were observed with increased pressure,despite an increase in complications. 展开更多

目的研究分析宝安区育龄妇女甲状腺功能的基本状况。方法 2015年1月-2017年6月期间宝安区下辖新安、西乡、福永、石岩、松岗以及沙井6个街道内15,000例育龄妇女作为此次研究的对象,根据所有的对象年龄将其分为高龄组和非高龄组。所有的... 目的研究分析宝安区育龄妇女甲状腺功能的基本状况。方法 2015年1月-2017年6月期间宝安区下辖新安、西乡、福永、石岩、松岗以及沙井6个街道内15,000例育龄妇女作为此次研究的对象,根据所有的对象年龄将其分为高龄组和非高龄组。所有的研究对象均在空腹状态下采集静脉血,对其TSH和FT4含量进行测定。分析所有研究对象的甲状腺功能状况。结果所有研究对象中甲状腺功能出现障碍的育龄妇女有995例,患病比例高达6.63%,其中甲状腺功能亢进的患者有596例,比例高达3.97%;甲状腺功能减退的患者有399例,占2.66%。在非高龄组和高龄组两组之间,高龄组中育龄妇女患有甲状腺功能亢进的患者有263例,比例高达9.28%,显著高于非高龄组,两组患者差异具有统计学意义(P<0.05);高龄组中育龄妇女患有甲状腺功能减退的患者有119例,患病比例为4.2%,与非高龄组相比,两组患者差异具有统计学意义(P<0.05)。所有正常研究对象中甲状腺功能出现亚甲亢和亚甲减的育龄妇女有1,756例,患病比例高达12.53%,其中亚甲亢的患者有995例,比例高达7.10%;亚甲减的患者有761例,占5.43%。在非高龄组和高龄组两组之间,高龄组中育龄妇女患有亚甲亢的患者有308例,比例高达12.56%,显著高于非高龄组,两组患者差异具有统计学意义(P<0.05);高龄组中育龄妇女患有亚甲减的患者有218例,患病比例为8.89%,与非高龄组相比,两组患者差异具有统计学意义(P<0.05)。结论宝安区育龄妇女的甲状腺功能存在着较高的异常率,特别是高龄妇女,应提高对其的关注和治疗程度,重视健康检查,降低出生缺陷,提高人口素质。 展开更多
