In recent years,with the continuous improvement and development of molecular technology in the application process,different types of DNA molecular markers have made rapid progress in the study of genetic diversity of rapeseed. The study of genetic diversity is conducive to the correct formulation of the strategy of collecting and in situ preservation of genetic resources of rapeseed,and it is the genetic basis for the improvement of rapeseed varieties. This article mainly starts with the three DNA molecular markers( SSR,RAPD,AFLP) widely used in the study of genetic diversity of rapeseed. By introducing the application principles and characteristics of SSR,RFPD and AFLP molecular markers,research progress of these three marker technologies in genetic diversity of rapeseed is briefly described. 展开更多

外周血淋巴细胞circRNA在脊柱结核患者中表达的初步筛选及功能分析 预览

目的:探寻对脊柱结核有诊断及治疗价值的特异性环状RNA(circRNA)及分析其功能。方法:选取2016年9月至2018年9月广西医科大学第一附属医院收治的25例脊柱结核患者作为实验组,25例健康志愿者作为对照组,并从中随机抽取3例脊柱结核患者和3... 目的:探寻对脊柱结核有诊断及治疗价值的特异性环状RNA(circRNA)及分析其功能。方法:选取2016年9月至2018年9月广西医科大学第一附属医院收治的25例脊柱结核患者作为实验组,25例健康志愿者作为对照组,并从中随机抽取3例脊柱结核患者和3例健康志愿者,分别提取外周血淋巴细胞中总RNA,进行circRNA基因芯片检测,筛选出脊柱结核患者和健康志愿者中差异表达的circRNAs,随后提取两组外周血淋巴细胞总RNA,并用RT-PCR方法验证所筛选的部分差异circRNA分子的表达情况。结果:通过circRNA基因芯片初步筛选出89个表达有差异的circRNAs,其中46个表达上调,43个表达下调。挑选在实验组中的上调和下调表达差异倍数最大的各5条circRNA进行RT-PCR验证,结果显示,与对照组相比,实验组中hsa_circRNA_100632、hsa_circRNA_103747、hsa_circRNA_102051、hsa_circRNA_104375、hsa_circRNA_405571表达上调,hsa_circRNA_104791、hsa_circRNA_400770、hsa_circRNA_403882、hsa_circRNA_104936、hsa_circRNA_404758表达下调,与基因芯片检测结果一致。结论:脊柱结核患者外周血淋巴细胞中存在一些特异性的circRNA表达,而且其有可能对脊柱结核的诊断及治疗有重要的意义。 展开更多

Insect body pigmentation and coloration are critical to adaption to the environment. To explore the mechanisms that drive pigmentation, we used the clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein 9 (Cas9) genome editing system to target the ebony gene in the non-model insect Spodoptera litura. Ebony is crucial to melanin synthesis in insects. By directly injecting Cas9 messenger RNA and ebony-specific guide RNAs into S. litura embryos, we successfully induced a typical ebony-deficient phenotype of deep coloration of the puparium and induction of melanin formation during the pupal stage. Polymerase chain reaction-based genotype analysis demonstrated that various mutations had occurred at the sites targeted in ebony. Our study clearly demonstrates the function of ebony in the puparium coloration and also provides a potentially useful marker gene for functional studies in S. litura as well as other lepidopteran pests. 展开更多

Application of Fingerprint Combined with QAMS in Quality Evaluation of Zhenrongdan Mixture 预览

[Objectives] The research aimed to evaluate the quality of Zhenrongdan mixture by fingerprint combining QAMS.[Methods] The quality evaluation method was established and validated with echinacoside as internal reference to determine the contents of other components(ferulic acid, salvianolic acid B, and icariin) according to the relative correction factor. The accuracy and feasibility of QAMS were evaluated by comparison on the results between the measured value and calculation value by external standard method and QAMS.[Results] A common pattern of characteristic fingerprint of Zhenrongdan mixture by HPLC was established. Thirteen common peaks were identified, and they account for 91% of the total peak area, and four components were verified in five batches of Zhenrongdan mixture. Good similarities with correlation coefficients higher than 0.99 were found in the fingerprints. There was no significant difference between the quantitative results of the four ingredients in the five batches by external standard method and QAMS.[Conclusions] The method of fingerprint combined with QAMS could be used for the quality control of multiple components determination and fingerprint chromatography for Zhenrongdan mixture. 展开更多

Recently, a novel green fluorescent protein eYGFPuv has been identified in the marine organism Chiridius poppei which displays high fluorescence intensity and can be visible by eyes in dark. Although strong green fluorescence was achieved in transgenic petunia, 3 expression cassettes (about 8 kb) complicate its application. In this study, to confirm whether 1 expression cassette could be used as a transgenic marker in prokaryotes and eukaryotes, eYGFPuv was cloned into prokaryotic expression vector pET28α-eYGFPuv- His and plant binary expression vector 35S::eYGFPuv. Compared to EGFP, eYGFPuv protein exhibited stronger dazzling green fluorescence in E. coli under excited light at 365 nm and maintains steadily over a long period of time without degradation. When transiently expressed in tobacco leaves, eYGFPuv protein displayed strong green fluorescence. Moreover, the fluorescence of eYGFPuv protein also could be directly observed in living plant, and thus can be used easily as a marker to screen transformed lines in transgenic research. Overall, compared to previous studies on eYGFPuv tandem repeats, our data confirmed that single eYGFPuv sequence still possesses high fluorescence intensity and quenching resistance. Furthermore, because of small size of expression cassette,it is suitable for efficient transformation in both prokaryotic and eukaryotic organisms. 展开更多

As an importantwild relative ofwheat,Agropyron cristatumhas been successfully used forwheat improvement.Currently,a few useful agronomic traits of A.cristatum,such as high grain number per spike and resistance to diseases,have been transferred into common wheat.However,the effective detection of small A.cristatum segmental introgressions in common wheat is still difficult.The objective of this study was to identify A.cristatum-specific single nucleotide polymorphisms (SNPs) for the detection of small alien segments in wheat.The transcriptome sequences of A.cristatum were aligned against wheat coding DNA sequences (CDS) for SNP calling.As a result,we discovered a total of 167,613 putative SNPs specific to the P genome of A.cristatum compared with the common wheat genomes.Among 230 selected SNPs with functional annotations related to inflorescence development and stress resistance,68 were validated as P genome-specific SNPs inmultiple wheat backgrounds using Kompetitive Allele Specific PCR (KASP) assays.Among them,55 SNPs were assigned to six homoeologous groups of the P genome using wheat-A.cristatum addition lines,and 6P-specific SNP markers were further physically mapped on different segments of chromosome 6P in 6P translocation lines.The P genome-specific SNPs were also validated by Sanger sequencing and used to detect the P chromatin in wheat-A.cristatum cryptic introgression lines.Two SNP markers (Unigene20217-182 and Unigene20307-1420) were detected in two wheat-A.cristatum introgression lines that showed enhanced grain number per spike and high resistance to powdery mildew.Together,thedeveloped P genome-specific SNPmarkers will accelerate thedetection of large numbers of wheat-A.cristatum derivatives andwill be helpful formarker-assisted transfer of desirable traits from A.cristatum into adapted wheat cultivars in wheat breeding programs. 展开更多

Cyst fluid glucose: An alternative to carcinoembryonic antigen for pancreatic mucinous cysts 预览

Pancreatic cystic lesions (PCLs) have been increasingly recognized in clinical practice. Although inflammatory cysts (pseudocysts) are the most common PCLs detected by cross-sectional imaging modalities in symptomatic patients in a setting of acute or chronic pancreatitis, incidental pancreatic cysts with no symptoms or history of pancreatitis are usually neoplastic cysts. For these lesions, it is imperative to identify mucinous cysts (intraductal papillary mucinous neoplasms and mucinous cystic neoplasms) due to the risk of their progression to malignancy. However, no single imaging modality alone is sufficient for a definitive diagnosis of all PCLs. The cyst fluid obtained by endoscopic ultrasound-guided fine needle aspiration provides additional information for the differential diagnosis of PCLs. Current recommendations suggest sending cyst fluid for cytology evaluation and measurement of carcinoembryonic antigen (CEA) levels. Unfortunately, the sensitivity of cytology is greatly limited, and cyst fluid CEA has demonstrated insufficient accuracy as a predictor of mucinous cysts. More recently, cyst fluid glucose has emerged as an alternative to CEA for distinguishing between mucinous and nonmucinous lesions. Herein, the clinical utility of cyst fluid glucose and CEA for the differential diagnosis of PCLs was evaluated. 展开更多

甲状腺癌病理分化类型与EB病毒感染及感染标志物水平的关联性分析

目的 分析甲状腺癌病理分化类型与EB病毒感染及感染标志物水平的关联性,阐明感染性疾病在恶性肿瘤发生和发展中的作用机制。方法 选取2017年2月-2018年2月80例医院收治的甲状腺癌患者作为研究组,选取同期80例甲状腺腺瘤患者作为对照组,... 目的 分析甲状腺癌病理分化类型与EB病毒感染及感染标志物水平的关联性,阐明感染性疾病在恶性肿瘤发生和发展中的作用机制。方法 选取2017年2月-2018年2月80例医院收治的甲状腺癌患者作为研究组,选取同期80例甲状腺腺瘤患者作为对照组,对两组患者的EB病毒感染率、血清白介素-6(IL-6)、C-反应蛋白(CRP)及外周血白细胞计数(WBC)水平进行观察和比较。结果 研究组患者EB病毒感染率及血清IL-6、CRP和WBC水平分别为46.25%、(0.56±0.18)μg/L、(5.26±0.71)mg/L、(7.65±2.82)×109/L,均高于对照组(P<0.05)。乳头状癌、滤泡状癌患者血清IL-6、CRP和WBC水平分别为(0.81±0.23)μg/L、(7.81±0.96)mg/L、(9.25±3.16)×109/L;(0.82±0.22)μg/L、(7.76±0.98)mg/L、(9.56±4.63)×109/L,高于髓样癌、未分化癌患者(P<0.05)。合并EB病毒感染甲状腺癌患者血清IL-6、CRP和WBC水平分别为(0.95±0.21)μg/L、(9.24±1.56)mg/L、(10.06±4.18)×109/L,均高于未合并EB感染患者(P<0.05)。结论 甲状腺癌患者的EB病毒感染率和感染标志物水平均高于良性甲状腺肿瘤患者,EB病毒感染可能与甲状腺癌患者的炎症反应具有关联性,某些病理分化类型甲状腺癌患者的EB病毒感染率较高,但尚未观察到与其他病毒类型存在显著的差异,还需要进一步的研究予以证实和讨论。 展开更多

Introduction:The histological diagnosis of prostate cancer is commonly based on morphological patterns.The presence of malignant tissue mixed with benign tissue,or the presence of carcinoma that mimics benignity may generate difficulty in the diagnostic elucidation.Therefore,the application of immunohistochemistry contributes its diagnostic value.Objectives:To evaluate the 34βE12 marker in the detection of adenocarcinoma(ADn),atypical small acinar proliferation(ASAp),regular prostatic tissue(RPT)and regular prostatic tissue alternated by atrophy spotlights(RPTa)in transrectal biopsy guided by ultrasonography of patients with suspected prostate cancer.Method:Analysis of 34 patients who underwent ultrasound-guided transrectal biopsy with subsequent analysis by H&E staining and 34βE12 labeling for elucidation of neoplasms or diseased tissues with doubtful diagnosis.Results:The marker 34βE12 showed negativity in 100%of the neoplasms ADn,positivity in 100%of the benign prostatic tissues(RPT and RPTa);the patients with ASAp presented positivity(20%)and negativity(80%).The chi-square test(χ2)showed that there is an association(χ2=29.55 and p<0.0001)between the groups,that is,the 34βE12 marker has a significant value(p<0.0001)in the elucidation of patients with prostatic neoplasia and benign prostatic tissues.Discussion and Conclusion:With the early screening of prostate cancer in the modern era,pathologists have become increasingly challenged to diagnose small outbreaks of cancer when only a few atypical glands are present in transrectal biopsy-guided ultrasonography.The 34βE12 marker becomes an important tool in elucidating diagnoses such as ADn and ASAp. 展开更多

Characterization of TaCOMT ge... Stem lignin content(SLC) in common wheat(Triticum aestivum L.) contributes to lodging resistance. Caffeic acid 3-O-methyltransferase(COMT) is a key enzyme involved in lignin biosynthesis. Characterization of TaCOMT genes and development of gene-specific markers could enable marker-assisted selection in wheat breeding. In the present study, the full-length genomic DNA(gDNA) sequences of TaCOMT genes located on chromosomes 3 A, 3 B, and 3 D were cloned by homologous cloning. Two allelic variants, TaCOMT-3 Ba and TaCOMT-3 Bb, were identified and differed by a 222-bp insertion/deletion(InDel) in the 3′-untranslated region(3′-UTR). A co-dominant gene-specific marker based on this InDel was developed and designated as Ta COMT-3 BM. A total of 157 wheat cultivars and advanced lines grown in four environments were used to validate the associations between allelic patterns and SLC. The SLC of cultivars with TaCOMT-3 Ba was significantly(P<0.01) higher than that of those with TaCOMT-3 Bb, and the marker TaCOMT-3 BM could be effectively used in wheat breeding. 展开更多

In this study, we employed RNA-seq technology to characterize transcriptomes for the flowers, leaves, and stems of this endangered herb. A total of 56 million clean reads were assembled into 120,716 unigenes with an N50 length of 850 bp. Among these unigenes, 70,245(58.19%) were successfully annotated and 65,965(54.64%) were identified as coding sequences based on their similarities with sequences in public databases. We identified 21 unigenes that had significant relationships with cold tolerance in N. incisum according to gene ontology(GO) annotation analysis. In addition, 13,149 simple sequence repeats(SSRs) and 85,681 single nucleotide polymorphisms were detected as potential molecular genetic markers. Ninety-six primer pairs of SSRs were randomly selected to validate their amplification efficiency and polymorphism. Nineteen SSR loci exhibited polymorphism in three natural populations of N. incisum. These results provide valuable resources to facilitate future functional genomics and conservation genetics studies of N. incisum. 展开更多

Starfruit (Averrhoa carambola L.) is an important fruit for Malaysian export and great attention has been made to improve starfruit fruit quality at Malaysian Agricultural Research and Development Institute (MARDI). The current study used next generation sequencing (NGS) technologies to develop starfruit simple sequence repeat (SSR) from 2 varieties namely B11 and B17 using Illumina HiSeq. The pre-processed reads were de novo assembled to generate approximately 75,000 and 74,000 scaffolds respectively. Total genome size for B11 and B17 were around 345 Mbp and 342 Mbp based on K-mer distribution analysis. In-silico microsatellite mining of each variety has identified more than 17,000 SSR in B11 and B17 respectively. Dinucleotides were the most abundant, accounting for more than 70% of all SSR and repeat motif GA (49%) was most common. A total of 239 SSR primer pairs were designed from contigs larger than 350 nucleotides and tested for amplification. The 30 polymorphic SSRs were used to DNA fingerprint of 12 starfruit hybrids. Polymorphism information content (PIC) ranged from 0.1411 to 0.6838, with an average of 0.3919. The Unweighted Pair-Group Method for Arithmetic Averages (UPGMA) dendrogram clustered 12 starfruit accessions into 2 groups. 展开更多

FUNAABOR-2 is a popular Ofada rice variety grown in a large area under rainfed upland condition across western states of Nigeria.We used the combination of phenotypic and marker-assisted selection(MAS)to improve grain yield of FUNAABOR-2 under drought stress(DS)at the reproductive stage via introgression of two drought quantitative trait loci(QTLs),qDTY12.1 and qDTY2.3.Foreground selection was carried out using peak markers RM511 and RM250,associated with qDTY12.1 and qDTY2.3,respectively,followed by recombinant selection with RM28099 and RM1261 distally flanking qDTY12.1.Furthermore,BC1F2-derived introgressed lines and their parents were evaluated under DS and non-stress(NS)conditions during the 2015–2016 dry season.Overall reduction of grain yield under DS compared to NS was recorded.Introgressed lines with qDTY12.1 and qDTY2.3 combinations showed higher yield potential compared to lines with single or no QTL under DS,indicating significant positive interactions between the two QTLs under the FUNAABOR-2 genetic background.Pyramiding of qDTY12.1 and qDTY2.3 in the FUNAABOR-2 genetic background led to higher grain yield production under DS and NS. 展开更多
