A biomolecule-free photoelectrochemical(PEC) probe(denoted as Ru-1) was designed, synthesized and coupled with TiO2 nanoparticles(NPs) for the highly sensitive and selective PEC detection of Hg^2+, a model analyte with hypertoxicity to both human health and ecosystem. The probe Ru-1 was designed with Ru(II) bipyridyl complex as the chromophore, thiocyanate ligand as the recognition unit, and carboxylate group as the linkage site to connect Ru-1 to TiO2 nanoparticles. Under irradiation, Ru-1 shows strong affinity to TiO2 and good photophysical properties of strong visible light absorption, slow electron-hole(e-h^+) recombination, and fast photoelectron injection to TiO2 nanoparticles via the bridge of 4-(2,2-bipyridin)-4-yl-thiophene moiety. However, the specific coordination of Hg^2+ with Ru-1 via the thiol moiety in the thiocyanate enlarges the band gap of tlie complex and reduces the photocurrent significantly. The synergistic interaction between TiO2 nanoparticles and the Ru-1 complex led to a selective PEC sensing strategy for Hg^2+ detection. Detectable linear ranges from 10^-12 g/mL to 10^-7 g/mL and from 10^-7g/mL to 10^-3 g/mL were obtained without the interference from possibly co-existed metal ions. The good analytical performances indicate the chemical probe based biomolecule-free PEC platform may offer a new route for the detection of biologically and environmentally important small molecules. 展开更多

Pt layers of 5 μm in thickness were electroplated before or after depositing NiCoCrAlY coating by arc ion plating(AIP) aiming for identifying the effect of Pt enriching position on microstructure and cyclic oxidation behavior of Pt modified NiCoCrAlY coatings. Al-rich zones formed at the same position of Ptrich zones for both modified coatings due to uphill diffusion of Al driven by Pt. Cyclic oxidation tests at 1000 and 1100?C indicated that oxidation resistance of NiCoCrAlY was improved by Pt modification via different mechanisms: at surface, Pt-rich zone promoted selective oxidation of Al to form α-Al2O3,whilst at coating/substrate interface Pt-rich zone acted as effective diffusion barrier for titanium. Roles of Pt played in enhancing the oxidation performance of various Pt-modified NiCoCrAlY coating were investigated. 展开更多

Herein, we reported a highly sensitive fluorescent probe DR, bearing a morpholine group and a BPEN ligand in the naphthalimide fluorophore. Upon Zn2+binding, DR exhibited remarkable fluorescence enhancement, and showed high sensitivity to Zn2+with the association constant of 4.9 ? 108 L/mol, and the detection limit of15 nmol/L. Confocal imaging experiments indicated that DR was able to localize to lysosomes in MCF-7 cells. Moreover, upon H2 O2 stimulation in SH-SY5 Y cells, endogenous release of Zn2+was observed. 展开更多

Bisulfite(HSO3) is an important sulfur dioxide(SO2) derivative, which plays a major role in many physiological processes and is also closely associated with a variety of diseases.Thus the development of highly selective and sensitive fluorescent probes is essential to detect HSO3 in living cells.In this work,we report the synthesis and analysis of a ratiometric fluorescent probe for selective detection of HSO3 based on the 1,4-nucleophilic addition reaction with the carbazole as an electron donor(D) and aldehyde group as an electron acceptor(A).The addition of HSO3 and other ions to our probe can be observed by UV vis and fluorescence spectrometry.Our investigation proved that the probe is highly selective and sensitive for HSO3 and ratiometric changes.Moreover, the probe has good cell permeability and was successfully applied to the detection of exogenous HSO3 in Hela cells and C.elegans. 展开更多

Carboxylesterase 1(CES1), one of the most abundant serine hydrolases in mammals, has drawn much attentions in recent years, owing to this enzyme involves in many physiological processes via hydrolysis of both endogenous esters and xenobiotic esters.Herein, to real-time monitor the activities of CES1 in various biological systems, a practical and iso form-specific fluorescent probe was developed on the basis of the substrate preference of CES1, as well as the structural and optical properties of BODIPY dyes.After screening of a panel of BODIPY ester derivatives, probe 1 displayed the best combination of specificity,sensitivity, enzymatic kinetics and applicability for monitoring CES1 activities in real samples.This probe was successfully used to detect CESl activities in several biological systems including tissue preparations,living cells, tissue slices and zebrafish.Furthermore, the biomedical applications of probe 1 for screening of CES1 inhibitors were also demonstrated using tissue preparations or living cells as enzyme sources.In summary, a practical and broadly applicable tool for real-time monitoring CES1 in biological systems was developed and well-characterized, which held great promise for further investigations on CES1-associated drug discovery, clinical practice and fundamental research. 展开更多

荧光原位杂交(Fluorescence in situ hybridization,FISH)在细胞遗传学中有较多应用,其中重复序列常被作为探针来研究物种进化、分类。但现常用的探针序列均较长(200 bp-500 bp),杂交耗时较多。本研究以烟草(Nictiana tabacum)为材料,... 荧光原位杂交(Fluorescence in situ hybridization,FISH)在细胞遗传学中有较多应用,其中重复序列常被作为探针来研究物种进化、分类。但现常用的探针序列均较长(200 bp-500 bp),杂交耗时较多。本研究以烟草(Nictiana tabacum)为材料,克隆并标记18S rDNA保守序列(254 bp),以其为探针对烟草染色体进行FISH,经18 h杂交获得了清晰的信号。在此基础上,利用荧光素直接标记引物序列(寡聚核苷酸),经较短时间(2 h)的杂交,也获得了清晰的信号。以寡聚核苷酸为探针对云烟87四倍体、N.tabacum-N.plumbaginifolia五倍体、烟草六倍体、云烟87八倍体以及N.plumbaginifolia的染色体进行杂交,均获得了良好效果。且杂交液可简化配制,洗脱过程也可简化。18S rDNA寡聚核苷酸探针与5S rDNA寡聚核苷酸探针结合,实现了N.plumbaginifolia的18S rDNA和5S rDNA双色FISH。所以,该寡聚核苷酸序列可以应用于烟草属植物18S rDNA(45S rDNA)位点的分析,且较大程度缩短了操作时间并简化了操作过程。 展开更多

An air-cooled probe was designed for measurement of total pressure at combustor outlet;its cooling scheme combined the film cooling and convection cooling.With the aid of CFD technique,cooling effectiveness of the coolant jets at various blow ratios was compared;suitable blow ratios and configuration of film holes were chosen accordingly.The overall cooling performance of the probe was evaluated via CFD technique,the design was improved according to the simulation result,and the cooling effect of the leading edge was obviously strengthened by increasing the local coolant mass flow rate.The results of wind tunnel test indicated that,between Mach numbers 0.2 and 0.4,the probe achieved a high accuracy at various attack angles.The probe was utilized in an annular combustor rig test,the highest temperature reached 1 760 Kand total pressure reaches 1 036 kPa.The result of rig test demonstrates that the coolant film distribution consistent appropriately with the CFD results. 展开更多

This study presents the Langmuir and Faraday probe measurements conducted to determine the plume characteristics of the BUSTLab microwave electrothermal thruster(MET).The thruster,designed to operate at 2.45 GHz frequency,is run with helium,argon and nitrogen gases as the propellant.For the measurements,the propellant volume flow rate and the delivered microwave power levels are varied.Experiments with nitrogen gas revealed certain operation regimes where a very luminous plume is observed.With the use of in-house-built Langmuir probes and a Faraday probe with guard ring,thruster plume electron temperature,plasma density and ion current density values are measured,and the results are presented.The measurements show that MET thruster plume effects on spacecraft will likely be similar to those of the arcjet plume.It is observed that the measured plume ion flux levels are very low for the high volume flow rates used for the operation of this thruster. 展开更多

Gold nanoclusters were rapid synthesized within 3 min at 120 ℃ by using papaya juice as a capping and reducing agent(P-AuNCs). The properties of the fluorescent probe were characterized by fluorescent spectroscopy, UV-vis spectroscopy, dynamic light scattering and transmission electron microscope.Based on the surface electron density increase-induced fluorescence enhancing principle, a high selective method for detection of L-lysine was developed with the as-prepared P-AuNCs coupling the fluorescence emission at 440 nm. The fluorescent probe showed high stability and good biocompatibility. Its fluorescence intensity was found to be linearly dependent on the L-lysine concentration in the range of 10.0μmol/L to 1000.0 μmol/L(R~2=0.969) with a limit of detection of 6.0μmol/L. Furthermore, the PAuNCs based approach was applied for monitoring the urine L-lysine contents, demonstrating great potential of fluorescent probes in real samples analysis. 展开更多

通过3,4-二甲氧基苯胺与2-硝基苯甲醛一步反应,合成了一种荧光增强型的硫化氢探针S2,并通过核磁和质谱确定了S2的结构。紫外分光光度计测试表明,探针S2与硫化钠发生了反应。在PBS缓冲液中,10μmol/L探针S2与硫化钠反应后,通过荧光分光... 通过3,4-二甲氧基苯胺与2-硝基苯甲醛一步反应,合成了一种荧光增强型的硫化氢探针S2,并通过核磁和质谱确定了S2的结构。紫外分光光度计测试表明,探针S2与硫化钠发生了反应。在PBS缓冲液中,10μmol/L探针S2与硫化钠反应后,通过荧光分光光度计进行荧光信号的检测。结果表明,探针S2可在8 min与硫化钠响应完全,随着硫化钠浓度的增加,探针S2在390 nm处的荧光值增加,且探针S2的荧光信号比值(F/F0)与硫化钠在0~8μmol/L的范围内呈线性关系,检出限为0. 20μmol/L。在与其他分析物的响应中,探针S2的选择性也很好。在自来水中通过加标回收实验,回收率在83. 3%~99. 4%之间。 展开更多
