OBJECTIVE:To investigate the therapeutic mechanism of compound Yindan decoction (CYD) in a rat model of acute intrahepatic cholestatic (AIC).METHODS:A total of 108 adult male rats were randomly divided into control (n =18) and AIC groups (n =90).AIC was induced in rats using alpha-naphthylisothiocyanate (ANIT)(75 mg/kg,10 mL/kg in corn oil,p.o.).Then,90 AIC rats were randomly divided into five groups:a control group (n =18),a CYD high dose group (n =18),a CYD middle dose group (n =18),a CYD low dose group (n =18),and a ursodeoxycholicacid (UDCA) group (n =18).According to sampling time,each group was subdivided into three subgroups:24 h (n =6),48 h (n =6),and 72 h groups (n =6).The CYD-high,-middle and-low groups were orally administered 24.48,12.24,and 6.12 g.kg-1.d-1 modified CYD,respectively,while the model group was given 20 mL/kg of body weight of distilled water once a day.The UDCA group was given 67.5 mg.kg-1.d-1 UDCA once a day.Radioimmunity assay was used to detect the activity of alanine aminotransferase (ALT),aspartate aminotransferase (AST),alkaline phosphatase (ALP),gamma-glutamyl transpeptidase (GGT) and the levels of total bilirubin (TBil) and indirectbiliruin (DBil) in rats.Reverse transcription quantitative polymerase chain reaction (qRT-PCR),Western blot analysis,and immunohistochemistry were used to detect multidrug resistance-associated protein 2 (MRP2) expression.In vitro,HepG2 hepatocellular carcinoma cells were treated with CYD medicated serum at a concentration of 15 mol/L.MRP2 and retinoid X receptor alpha (RXRα) expression was analyzed by qRT-PCR and Western blotting.RESULTS:Serum levels of ALT,AST,GGT,ALP,TBil,and DBil were significantly reduced in the CYD and positive drug groups compared with the control group (P < 0.05 and P < 0.01,respectively).Pathological changes in rat liver tissues at 72 h in the CYD-high and-medium dose groups and positive drug group were not significant compared with the control group.CYD and UDCA treatment ameliorated ANIT-induced biliary epithelial cell pro 展开更多

OBJECTIVE:To evaluate the effect of Wulong Xiaozheng Wan medicated serum on the epithelial-mesenchymal transition (EMT) of BGC823 cell induced by transforming growth factor-β,(TGF-β,) and to explore its mechanism.METHODS:EMT model of BGC823 was stimulated by TGF-β1.Wulong Xiaozheng Wan medicated serum and LY-364947 were used as intervention.The proliferation and adhesion of BGC823 were detected by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide and flow cytometry was used to detect the apoptosis.The invasion and migration were detected by Transwell.The level of matrix metalloproteins was detected by enzyme-linked immunosorbent assay.The expressions of related proteins and mRNA of EMT marker and TGF-β1/Smad signal pathway were detected by Western blot and reverse transcription-polymerase chain reaction.RESULTS:Compared with the TGF-β1 group,Wulong Xiaozheng Wan medicated serum could inhibit the ability of proliferation,heterogeneous adhesion,invasion,and migration.It also promotes apoptosis and homotypic adhesion in BGC823,with a dose-dependent manner.Meanwhile,Wulong Xiaozheng Wan medicated serum could regulate the expression of related proteins and mRNA of TGF-β1/Smad signaling pathway,and inhibit the expressions of EMT transcription factors and EMT markers.CONCLUSION:Wulong Xiaozheng Wan medicated serum inhibited epithelial-mesenchymal transition by down-regulated the expression of TβRI and the activation of TGF-β1/Smad signaling pathway. 展开更多

Late embryogenesis abundant (LEA) proteins accumulate in the late stage of plant seed development, and are upregulated in most plants during drought, cold, heat, or salinity stress. LEA proteins can be classified by amino-acid sequence into seven groups. Dehydrins belong to LEA protein group Ⅱ. In previous studies, the maize KS type dehydrin ZmDHN13 increased the tolerance of transgenic tobacco to oxidative stress. In the present study, ZmDHN13 was identified under copper stress conditions, and the protein was then characterized using transgenic yeast and tobacco plants to investigate its functions. ZmDHN13 bound Cu2+. Its overexpression in transgenic tobacco conferred tolerance to copper stress by binding metals and reducing the accumulation of reactive oxygen species (ROS). Three conserved domains displayed a cooperative effect under copper stress conditions. 展开更多

维持性血液透析患者血清klotho蛋白水平与营养状况的关系

目的探究维持性血液透析(MHD)患者血清klotho蛋白水平与营养状况的关系。方法选取本院2016年6月至2016年11月治疗的维持性血液透析患者100例作为研究组,根据主观综合性营养评估(SGA)法将研究组分为三组:A组:营养良好组28例,B组:轻中度... 目的探究维持性血液透析(MHD)患者血清klotho蛋白水平与营养状况的关系。方法选取本院2016年6月至2016年11月治疗的维持性血液透析患者100例作为研究组,根据主观综合性营养评估(SGA)法将研究组分为三组:A组:营养良好组28例,B组:轻中度营养不良组44例,C组:重度营养不良组28例,并选取同期健康体检者60例作为对照组,采用微主观综合性营养评估(SGA)法评价两组患者营养的状况,采用ELISA法测定两组对象血清klotho蛋白水平,并对两组患者相关生化指标测定。利用Pearson相关分析法分析血清klotho蛋白水平与相关生化指标的相关性。结果研究组患者合并营养不良的发生率为72%(72/100),明显高于对照组的18.32%(11/60)(P<0.05);且不同营养状况A、B、C三组血清各指标水平存在组间差异(P<0.05),与A组相比较,B组及C组患者BMI、ALB、Hb、MAMC、klotho蛋白水平均降低,PTH、FGF-23升高,差异有统计学意义(P<0.05);与B组相比,C组患者Scr、ALB、Hb、MAMC、klotho蛋白水平均降低,PTH、FGF-23升高,差异有统计学意义(P<0.05);血清klotho蛋白水平与ALB、MAMC、Hb呈正相关(P<0.05);与BMI无相关性(P=0.165);与Scr、BUN、FGF23、PTH呈负相关(P<0.05)。结论维持性血液透析患者合并营养不良的发生率很高,血清klotho蛋白水平的低表达与患者营养状况密切相关,为尿毒症患者合并营养不良的诊断、治疗提供了新依据。 展开更多

Antibiotics have been widely applied as effective agents for curing infectious diseases caused by pathogenic bacteria. However, the emergence of antibiotic resistance shows a growing threat to the effectiveness of clinical applications of antibiotics. To overcome this challenge, identifying new promising targets for drug development is now urgently needed. The inhibition of translation process is one of the most important mechanisms for many of the existing antibiotics. Meanwhile, ribosome is a universal translational machine in all living cells. It can link amino acids together through the guiding of mRNAs, functioning as the workplace of protein synthesis or translation. Targeting the regulation of ribosomal function and ribosome assembly is an attracting solution for identifying novel targets for antibiotic development. These targets are very critical for developing antibiotics with new mode of action, thus, holds the promise for tackling the ever-growing crisis of bacterial drug resistance. 展开更多

Plant peroxisomes are unique subcellular organelles which play an indispensable role in several key metabolic pathways, including fatty acid b-oxidation,photorespiration, and degradation of reactive oxygen species. The compartmentalization of metabolic pathways into peroxisomes is a strategy for organizing the metabolic network and improving pathway efficiency. An important prerequisite, however, is the exchange of metabolites between peroxisomes and other cell compartments. Since the first studies in the 1970s scientists contributed to understanding how solutes enter or leave this organelle.This review gives an overview about our current knowledge of the solute permeability of peroxisomal membranes described in plants, yeast, mammals and other eukaryotes. In general, peroxisomes contain in their bilayer membrane specific transporters for hydrophobic fatty acids(ABC transporter) and large cofactor molecules(carrier for ATP, NAD and CoA). Smaller solutes with molecular masses below 300–400 Da, like the organic acids malate, oxaloacetate, and 2-oxoglutarate, are shuttled via non-selective channels across the peroxisomal membrane.In comparison to yeast, human, mammals and other eukaryotes, the function of these known peroxisomal transporters and channels in plants are discussed in this review. 展开更多

In this study,we revealed the differential proteins from the wheat endosperms using proteomic analysis and investigated their surface properties.The pattern of the polypeptides obtained from the Yangmai-15 and Yangmai-16 wheat varieties were compared using two-dimensional polyacrylamide gels.In addition,we compared the characteristics of the grain such as grain hardness,protein content,wet gluten,dough development time,dough stability,gliadin and glutenin contents between Yangmai-15 and Yangmai-16,and the results were significantly different.Notably,216 and 197 protein spots were separated from Yangmai-15 and Yangmai-16,respectively.The isoelectric points of the identified proteins ranged from 4 to 10 and the molecular weights of proteins varied from 10 to 100 kDa.Further,21 and 8 specific differential protein spots were identified fromthe flour of Yangmai-15 and Yangmai-16,respectively.The surface properties of identified peptides consisted of hydrophobic or hydrophilic residues,as well as randomly scattered residues.The proteomic analysis of the wheat endosperms provides a novel insight into the biochemical basis for the differences in physicochemical properties between the soft and hard wheat varieties. 展开更多

Analyzing proteins in colonic tissues from mice with ulcerative colitis using the iTRAQ technology 预览

Objective The aim of the study was to investigate the expression of proteins in colonic tissues of mice with ulcerative colitis(UC) by using isobaric tags for relative and absolute quantitation(iTRAQ), probe into the pathogenesis of UC, and find potential biomarkers of UC. Methods Forty C57 mice were randomly divided into the control and model groups(20 mice in each group). The mice in the model group were administered dextran sulphate sodium(DSS) for 7 consecutive days ad libitum to induce acute colitis, and the colon tissue was extracted on the 8 th day after the successful establishment of the UC model. Proteins were identified by the i TRAQ and tandem mass spectrometry techniques,and the identified proteins were analyzed by bioinformatics. Results A total of 4019 proteins were identified among the two groups. Among them, 317 significant differentially expressed proteins(DEPs) were detected according to the screening criteria for selecting DEPs, i.e. fold change ratios ≥ 1.5 or ≤ 0.67 and P-values < 0.05, of which 156 were upregulated and 161 were downregulated. In the Gene Ontology(GO) analysis, the DEPs were classified into 48 functional categories, which contained biological process, cellular component, and molecular function. Based on the 317 DEPs, the KEGG pathway analysis identified 160 vital pathways.Conclusion DEPs in colonic tissues of mice with UC were screened using the iTRAQ technique, which laid a foundation for further studies regarding the pathogenesis of UC. 展开更多

In semi-arid region of northwestern China, underground mining subsidence often results in decreased vegetation coverage, impoverishment of soil fertility and water stress. In addition, the physical-chemical and biological properties of soil also change, resulting in more susceptible to degradation. In particular, subsidence causes disturbance of the symbioses of plant and microbe that can play a beneficial role in the establishment of vegetation communities in degraded ecosystems. The objective of this study was to evaluate the effects of revegetation with exotic arbuscular mycorrhizal fungi(AMF) inoculum on the chemical and biological properties of soil over time in mining subsidence areas. Soils were sampled at a depth up to 30 cm in the adjacent rhizosphere of Amorpha fruticose Linn. from five reclaimed vegetation communities in northwestern China. In August 2015, a field trial was set up with five historical revegetation experiments established in 2008(7-year), 2011(4-year), 2012(3-year), 2013(2-year) and 2014(1-year), respectively. Each reclamation experiment included two treatments, i.e., revegetation with exotic AMF inoculum(AMF) and non-AMF inoculum(the control). Root mycorrhizal colonization, glomalin-related soil protein(GRSP), soil organic carbon(SOC), soil nutrients, and enzyme activities were also assessed. The results showed that mycorrhizal colonization of inoculated plants increased by 33.3%–163.0% compared to that of non-inoculated plants(P<0.05). Revegetation with exotic AMF inoculum also significantly improved total GRSR(T-GRSP) and easily extracted GRSP(EE-GRSP) concentrations compared to control, besides the T-GRSP in 1-year experiment and the EE-GRSP in 2-year experiment. A significant increase in SOC content was only observed in 7-year AMF reclaimed soils compared to non-AMF reclaimed soils. Soil total N(TN), Olsen phosphorus(P) and available potassium(K) were significantly higher in inoculated soil after 1–7 years of reclamation(except for individual cases), and increased with reclamat 展开更多

【目的】探讨围术期血红蛋白(Hb)与凝血酶原时间(PT)监测评估在老年主动脉瓣狭窄患者主动脉瓣置换术后发生消化道出血风险的临床价值。【方法】选取2016年5月至2017年9月本院收治的76例行主动脉瓣置换术治疗的主动脉瓣狭窄患者临床资料... 【目的】探讨围术期血红蛋白(Hb)与凝血酶原时间(PT)监测评估在老年主动脉瓣狭窄患者主动脉瓣置换术后发生消化道出血风险的临床价值。【方法】选取2016年5月至2017年9月本院收治的76例行主动脉瓣置换术治疗的主动脉瓣狭窄患者临床资料,根据术后是否出现消化道出血将其分为观察组(n =16)和对照组(n =60)。收集患者术前12 h(T0)、术中(T1)、术后12 h(T2)三个不同时段的血液标本,其中Hb以全血为样本,采集后立即检测,检测方法为高效液相色谱法;凝血酶原时间(PT)采用SysmexCA-7000全自动血凝分析仪进行测定。【结果】两组患者T1时Hb水平显著低于T0时,T2时显著高于T1时,观察组T1、T2时Hb水平显著低于对照组,差异均有统计学意义(P <0.05)。两组患者T1时PT大于T0时,T2时刻PT小于T1时,观察组T1、T2时PT大于对照组,差异均有统计学意义(P <0.05)。观察患者T0、T1、T2三个不同时段Hb与PT水平均呈负相关(P <0.05);对照组患者T1时Hb与PT水平呈负相关(P <0.05),T0 、T2时Hb与PT水平无相关性(P >0.05)。【结论】围术期Hb与PT水平监测对于降低老年主动脉瓣狭窄患者主动脉瓣置换术后消化道出血风险的有一定的提示作用。在检测过程中应重点关注Hb水平降低与PT水平升高的患者,一旦出现相关的术后并发症应及时进行病因和对症处理。 展开更多

【目的】分析脑梗死合并高脂血症患者血管内皮生长因子(vascular endothelial growth factor, VEGF)、血浆脂蛋白相关磷脂A2(Plasma lipoprotein associated phospholipid A2, Lp-PLA2)及血小板活化功能的变化及意义。【方法】随机选取2... 【目的】分析脑梗死合并高脂血症患者血管内皮生长因子(vascular endothelial growth factor, VEGF)、血浆脂蛋白相关磷脂A2(Plasma lipoprotein associated phospholipid A2, Lp-PLA2)及血小板活化功能的变化及意义。【方法】随机选取2017年1月至2018年9月本院收治的脑梗死合并高脂血症患者56例作为观察组,同期选择健康体检者30例作为对照组。采用ELISA法检测所有受试者血清VEGF及Lp-PLA2水平,流式细胞仪检测血小板活化功能指标;同时采用SPSS Pearson相关性分析软件对VEGF、Lp-PLA2水平及血小板活化功能指标与脑梗死合并高脂血症的关系进行分析。【结果】观察组血清VEGF、血浆Lp-PLA2、血小板-单核细胞聚集体(Platelet-monocyte aggregates, PM A)、溶酶体颗粒膜糖蛋白(CD63)及血小板上a颗粒膜糖蛋白(CD62p)均显著高于对照组,差异具有统计学意义(P<0.05)。Pearson相关性分析显示,VEGF、Lp-PLA2水平及血小板活化功能指标与脑梗死合并高脂血症呈正相关(P<0.05)。【结论】脑梗死合并高脂血症患者血小板活化指标表达升高,脑组织存在缺氧及炎性浸润状态,治疗时候需抑制血小板活化、改善缺氧同时控制炎性反应。 展开更多

【目的】探究AKIP1对人结直肠癌细胞HCT-116凋亡的影响及其相关分子机制。【方法】设计AKIP1干扰片段siRNA及NC片段,转染至HCT-116细胞,Real-time PCR及Western Blot检测AKIP1 mRNA及蛋白的表达;通过MTT及流式细胞术检测细胞活力及凋亡... 【目的】探究AKIP1对人结直肠癌细胞HCT-116凋亡的影响及其相关分子机制。【方法】设计AKIP1干扰片段siRNA及NC片段,转染至HCT-116细胞,Real-time PCR及Western Blot检测AKIP1 mRNA及蛋白的表达;通过MTT及流式细胞术检测细胞活力及凋亡情况;通过Real-time PCR检测细胞凋亡相关基因的表达变化,并通过Western Blot检测NF-κB p65在胞浆和胞核中的表达以及凋亡相关蛋白的表达。【结果】转染AKIP1 siRNA后,AKIP1 mRNA及蛋白的表达水平显著下调;MTT实验结果显示AKIP1 siRNA可显著抑制HCT-116细胞的活力;流式细胞术结果表明AKIP1 siRNA可显著促进HCT-116细胞凋亡;同时干扰AKIP1可上调Bax、cleaved-Caspase-3并下调Bcl-2的表达;另外,AKIP1 siRNA可下调细胞核NF-κB p65的表达,上调细胞浆中的NF-κB p65蛋白表达。【结论】AKIP1可能通过活化NF-κB p65抑制HCT-116细胞凋亡进而促进结直肠癌的疾病进展。 展开更多

近年来国内外对人乳成分的床旁快速测定趋于增多。国外大部分应用红外色谱技术,其中中红外线技术在近年的应用更为普遍。近年的系统综述显示,红外色谱技术对蛋白质和脂肪测定的结果准确性为60% 70%;对碳水化合物和乳糖的测定结果总体不... 近年来国内外对人乳成分的床旁快速测定趋于增多。国外大部分应用红外色谱技术,其中中红外线技术在近年的应用更为普遍。近年的系统综述显示,红外色谱技术对蛋白质和脂肪测定的结果准确性为60% 70%;对碳水化合物和乳糖的测定结果总体不肯定。国内普遍应用的数字化超声测定人乳成分仅有个别小样本报告,因而难以评估真正的准确性。测定结果同时受到人乳样本的保存方法、温度和盛放容器等条件的影响。人乳营养素含量的差异和变动,包括个体人乳在哺乳期不同时间的差别也造成测定结果不稳定,从而缺乏完整的代表性。基于人乳快速测定的个体化强化可减少强化剂使用的偏差,然而对早产儿生长速率无显著影响。因此,人乳成分测定在临床应用目前缺乏证据,也不宜常规开展。 展开更多

目的通过分析多索茶碱对慢性阻塞性肺疾病(COPD)急性加重期的疗效及对JAK/STAT通路蛋白表达的影响,为临床治疗提供参考。方法选取2015年1月—2017年7月琼海市中医院收治的COPD急性加重期患者,分析患者多索茶碱的疗效及治疗前后血清中JAK... 目的通过分析多索茶碱对慢性阻塞性肺疾病(COPD)急性加重期的疗效及对JAK/STAT通路蛋白表达的影响,为临床治疗提供参考。方法选取2015年1月—2017年7月琼海市中医院收治的COPD急性加重期患者,分析患者多索茶碱的疗效及治疗前后血清中JAK/STAT通路蛋白的表达变化。结果COPD急性加重期患者JAK1、JAK2、STAT1和STAT2蛋白相对表达量较对照组高(P<0.05);COPD急性加重期组治疗后FEV1、FVC、FEV1/FVC及PaO2较治疗前升高(P<0.05),PaCO2较治疗前降低(P<0.05);COPD急性加重期患者治疗前JAK1、JAK2、STAT1和STAT2蛋白相对表达量高于治疗后(P<0.05)。结论多索茶碱对COPD急性加重期患者肺功能有明显的改善作用,且对JAK/STAT通路蛋白表达有明显的下调作用。 展开更多
