Stem radial CO2 conductance affects stem respiratory CO2 fluxes in ash and birch trees 预览

The CO2 released from respiring cells in woody tissues of trees can contribute to one of three fluxes:efflux to the atmosphere(EA),internal xylem sap transport flux(FT),and storage flux(△S).Adding those fluxes together provides an estimate of actual stem respiration(RS).We know that the relative proportion of CO2 in those fluxes varies greatly among tree species,but we do not yet have a clear understanding of the causes for this variation.One possible explanation is that species differ in stem radial CO2 conductance(gc).A high gc would favor the EA pathway and a low gc would favor the FT pathway.However,gc has only been measured once in situ and only in a single tree species.We measured gc using two methods in stems of Fraxinus mandshurica Rupr.(ash)and Betula platyphylla Suk.(birch)trees in situ,along with RS,EA,FT and △S.Stem radial CO2 conductance was substantially greater in ash trees than in birch trees.Corresponding to that finding,in ash trees over 24 h,EA constituted the entire flux of respired CO2,and FT was negative,indicating that additional CO2,probably transported from the root system via the xylem,was also diffusing into the atmosphere.In ash trees,FT was negative over the entire 24 h,and this study represents the first time that has been reported.The addition of xylem-transported CO2 to EA caused EA to be 9%higher than the actual RS over the diel measurement period.Birch trees,which had lower gc,also had a more commonly seen pattern,with EA accounting for about 80%of the CO2 released from local cell respiration and FT accounting for the remainder.The inorganic carbon concentration in xylem sap was also lower in ash trees than in birch trees:2.7 versus 5.3 mmol L^-1,respectively.Our results indicate that stem CO2 conductance could be a very useful measurement to help explain differences among species in the proportion of respired CO2that remains in the xylem or diffuses into the atmosphere. 展开更多

Currently,there does not exist a strategy that can reduce diabetes and scientists are working towards a cure and innovative approaches by employing stem cellbased therapies.On the other hand,bioprinting technology is a novel therapeutic approach that aims to replace the diseased or lostβ-cells,insulin-secreting cells in the pancreas,which can potentially regenerate damaged organs such as the pancreas.Stem cells have the ability to differentiate into various cell lines including insulinproducing cells.However,there are still barriers that hamper the successful differentiation of stem cells intoβ-cells.In this review,we focus on the potential applications of stem cell research and bioprinting that may be targeted towards replacing theβ-cells in the pancreas and may offer approaches towards treatment of diabetes.This review emphasizes on the applicability of employing both stem cells and other cells in 3D bioprinting to generate substitutes for diseasedβ-cells and recover lost pancreatic functions.The article then proceeds to discuss the overall research done in the field of stem cell-based bioprinting and provides future directions for improving the same for potential applications in diabetic research. 展开更多

Pluripotent stem cells(PSCs) are characterized by their capacity for high self-renewal and multiple differentiation potential and include embryonic stem cells, embryonic germ cells and induced PSCs. PSCs provide a very suitable model for the studies of human diseases, drugs screening, regenerative medicine and developmental biology research. Pigs are considered as an ideal model for preclinical development of human xenotransplantation, therapeutic approaches and regenerative medicine because of their size and physiological similarity to humans. However, lack of knowledge about the derivation, characterization and pluripotency mechanisms of porcine PSCs hinders progress in these biotechnologies. In this review, we discuss the latest progress on porcine PSCs generation, evaluation criteria for pluripotency, the scienti?c and technical questions arising from these studies. We also introduce our perspectives on porcine PSC research, in the hope of providing new ideas for generating naive porcine PSCs and animal breeding. 展开更多

Induced pluripotent stem cells(iPSCs)were first generated by Yamanaka and colleagues over a decade ago.Since then,iPSCs have been successfully differentiated into many distinct cell types,enabling tissue-,disease-,and patientspecific in vitro modelling.Cardiovascular disease is the greatest cause of mortality worldwide but encompasses rarer disorders of conduction and myocardial function for which a cellular model of study is ideal.Although methods to differentiate iPSCs into beating cardiomyocytes(iPSC-CMs)have recently been adequately optimized and commercialized,the resulting cells remain largely immature with regards to their structure and function,demonstrating fetal gene expression,disorganized morphology,reliance on predominantly glycolytic metabolism and contractile characteristics that differ from those of adult cardiomyocytes.As such,disease modelling using iPSC-CMs may be inaccurate and of limited utility.However,this limitation is widely recognized,and numerous groups have made substantial progress in addressing this problem.This review highlights successful methods that have been developed for the maturation of human iPSC-CMs using small molecules,environmental manipulation and 3-dimensional(3D)growth approaches. 展开更多

Stem cell transplantation has brought new hope for the treatment of neurological diseases.The key to stem cell therapy lies in inducing the specific differentiation of stem cells into nerve cells.Because the differentiation of stem cells in vitro and in vivo is affected by multiple factors,the final differentiation outcome is strongly associated with the microenvironment in which the stem cells are located.Accordingly,the optimal microenvironment for inducing stem cell differentiation is a hot topic.EGb761 is extracted from the leaves of the Ginkgo biloba tree.It is used worldwide and is becoming one of the focuses of stem cell research.Studies have shown that EGb761 can antagonize oxygen free radicals,stabilize cell membranes,promote neurogenesis and synaptogenesis,increase the level of brain-derived neurotrophic factors,and replicate the environment required during the differentiation of stem cells into nerve cells.This offers the possibility of using EGb761 to induce the differentiation of stem cells,facilitating stem cell transplantation.To provide a comprehensive reference for the future application of EGb761 in stem cell therapy,we reviewed studies investigating the influence of EGb761 on stem cells.These started with the composition and neuropharmacology of EGb761,and eventually led to the finding that EGb761 and some of its important components play important roles in the differentiation of stem cells and the protection of a beneficial microenvironment for stem cell transplantation. 展开更多

Osteoarthritis (OA) refers to a chronic joint disease characterized by degenerative changes of articular cartilage and secondary bone hyperplasia. Since articular cartilage has a special structure, namely the absence of blood vessels as well as the low conversion rate of chondrocytes in the cartilage matrix, the treatment faces numerous clinical challenges. Traditional OA treatment (e.g., arthroscopic debridement, microfracture, autologous or allogeneic cartilage transplantation, chondrocyte transplantation) is primarily symptomatic treatment and pain management, which cannot contribute to regenerating degenerated cartilage or reducing joint inflammation. Also, the generated mixed fibrous cartilage tissue is not the same as natural hyaline cartilage. Mesenchymal stem cells (MSCs) have turned into the most extensively explored new therapeutic drugs in cell-based OA treatment as a result of their ability to differentiate into chondrocytes and their immunomodulatory properties. In this study, the preliminary results of preclinical (OA animal model)/clinical trials regarding the effects of MSCs on cartilage repair of knee joints are briefly summarized, which lay a solid application basis for more and deeper clinical studies on cell-based OA treatment. 展开更多

Effect of adipose-derived mesenchymal stem cells on hepatocellular carcinoma:In vitro inhibition of carcinogenesis

AIM To investigate the effect of adipose-derived mesenchymal stem cells(ADMSCs)and their conditioned media(CM)on hepatocellular carcinoma(HCC)cell tumorigenesis.METHODS The proliferation rate of HepG2 and PLC-PRF-5 HCC cancer cells was measured using the trypan blue exclusion method and confirmed using the cell-counting kit 8(commonly known as CCK-8)assay.Apoptosis was detected by flow cytometry using annexin V-FITC.Protein and mRNA expression was quantified by ELISA and real time PCR,respectively.Migration and invasion rates were performed by Transwell migration and invasion assays.Wound healing was examined to confirm the data obtained from the migration assays.RESULTS Our data demonstrated that when co-culturing HCC cell lines with ADMSCs or treating them with ADMSC CM,the HCC cell proliferation rate was significantly inhibited and the apoptosis rate increased.The decreased proliferation rate was accompanied by an upregulation of P53 and Retinoblastoma mRNA and a downregulation of c-Myc and hTERT mRNA levels.More notably,ADMSCs and their CM suppressed the expression of the two important markers of HCC carcinogenicity,alpha-fetoprotein and Des-gamma-carboxyprothrombin.In addition,the migration and invasion levels of HepG2 and PLC-PRF-5 cells significantly decreased,potentially through increased expression of the tissue inhibitor metalloproteinases TIMP-1,TIMP-2 and TIMP-3.CONCLUSION These findings shed new light on a protective and therapeutic role for ADMSCs and their CM in controlling HCC invasiveness and carcinogenesis. 展开更多

Cellular reprogramming and induced pluripotent stem cell(IPSC)technology demonstrated the plasticity of adult cell fate,opening a new era of cellular modelling and introducing a versatile therapeutic tool for regenerative medicine.While IPSCs are already involved in clinical trials for various regenerative purposes,critical questions concerning their medium-and long-term genetic and epigenetic stability still need to be answered.Pluripotent stem cells have been described in the last decades in various mammalian and human tissues(such as bone marrow,blood and adipose tissue).We briefly describe the characteristics of human-derived adult stem cells displaying in vitro and/or in vivo pluripotency while highlighting that the common denominators of their isolation or occurrence within tissue are represented by extreme cellular stress.Spontaneous cellular reprogramming as a survival mechanism favoured by senescence and cellular scarcity could represent an adaptative mechanism.Reprogrammed cells could initiate tissue regeneration or tumour formation dependent on the microenvironment characteristics.Systems biology approaches and lineage tracing within living tissues can be used to clarify the origin of adult pluripotent stem cells and their significance for regeneration and disease. 展开更多

BACKGROUND Pluripotent stem cell-derived cardiomyocytes(CMs)have become one of the most attractive cellular resources for cell-based therapy to rescue damaged cardiac tissue.AIM We investigated the regenerative potential of mouse embryonic stem cell(ESC)-derived platelet-derived growth factor receptor-α(PDGFRα)^+cardiac lineagecommitted cells(CLCs),which have a proliferative capacity but are in a morphologically and functionally immature state compared with differentiated CMs.METHODS We induced mouse ESCs into PDGFRα^+CLCs andαMHC^+CMs using a combination of the small molecule cyclosporin A,the rho-associated coiled-coil kinase inhibitor Y27632,the antioxidant Trolox,and the ALK5 inhibitor EW7197.We implanted PDGFRα^+CLCs and differentiatedαMHC+^CMs into a myocardial infarction(MI)murine model and performed functional analysis using transthoracic echocardiography(TTE)and histologic analysis.RESULTS Compared with the untreated MI hearts,the anterior and septal regional wall motion and systolic functional parameters were notably and similarly improved in the MI hearts implanted with PDGFRα^+CLCs andαMHC+CMs based on TTE.In histologic analysis,the untreated MI hearts contained a thinner ventricular wall than did the controls,while the ventricular walls of MI hearts implanted with PDGFRα^+CLCs andαMHC^+CMs were similarly thicker compared with that of the untreated MI hearts.Furthermore,implanted PDGFRα+CLCs aligned and integrated with host CMs and were mostly differentiated intoα-actinin^+CMs,and they did not convert into CD31^+end othelial cells orαSMA^+mural cells.CONCLUSION PDGFRα^+CLCs from mouse ESCs exhibiting proliferative capacity showed a regenerative effect in infarcted myocardium.Therefore,mouse ESC-derived PDGFRα^+CLCs may represent a potential cellular resource for cardiac regeneration. 展开更多

Animal experiments have confirmed that mesenchymal stem cells can inhibit motor neuron apoptosis and inflammatory factor expression and increase neurotrophic factor expression. Therefore, mesenchymal stem cells have been shown to exhibit prospects in the treatment of amyotrophic lateral sclerosis. However, the safety of their clinical application needs to be validated. To investigate the safety of intrathecal injection of Wharton's jelly-derived mesenchymal stem cells in amyotrophic lateral sclerosis therapy, 43 patients (16 females and 27 males, mean age of 57.3 years) received an average dose of 0.42 × 106 cells/kg through intrathecal administration at the cervical, thoracic or lumbar region depending on the clinical symptoms. There was a 2 month interval between two injections. The adverse events occurring during a 6-month treatment period were evaluated. No adverse events occurred. Headache occurred in one case only after first injection of stem cells. This suggests that intrathecal injection of Wharton's Jelly-derived mesenchymal stem cells is well tolerated in patients with amyotrophic lateral sclerosis. This study was approved by the Bioethical Committee of School of Medicine, University of Warmia and Mazury in Olsztyn, Poland (approval No. 36/2014 and approval No. 8/2016).
