Exogenous ligands include... A capture strategy is described and demonstrated for retrieving ligand entities in plasma that bind Human Serum Albumin. The method has applications for both exogenous and endogenous ligands. Exogenous ligands include drug candidates, performance enhancing drugs and toxic nerve agents that also interact quite strongly with HSA. Endogenous ligands are natural circulating compounds whose abundance corresponds to normal hemostasis or elevated levels that could be disease-specific molecular biomarkers. Melting curves of plasma solutions measured by differential scanning calorimetry produce “so-called” plasma thermograms that are physical signatures of the plasma solution. Patterns displayed by thermograms can be sensitive indicators of the presence of abnormal levels of exogenous and endogenous ligand components. Effects of ligand interactions on thermodynamic stability of proteins in plasma that they bind, primarily HSA, manifest on the plasma thermogram. The capture strategy is demonstrated for HSA binding in plasma of four “ideal” ligands of different types. The particular ligands were naproxen, bromocresol green, short double stranded and single strand DNA. Thermogram shapes and features were sensitive to the presence of ligands as thermograms of mixtures of plasma and HSA with these ligands were significantly different than thermograms of plasma or HSA alone. These results demonstrated directly that significant perturbations of plasma thermograms corresponded to ligand interactions with HSA in plasma. 展开更多

Cascadia Convergent Zone: An Example of Primary Convergent Seismogenic Structure 预览

In this article, a case is made for very-large or primary seismogenic structures in convergent margins, based on anomalous large earthquake magnitudes (Mw 8 - 9) relative to rupture lengths. Out of 56,293 earthquakes (magnitudes ≥ 5) cataloged worldwide, the 10 largest events in transform, divergent, and interior settings average magnitudes of 7.3 - 7.6. But in convergent margins, the average magnitude of the 10 largest events is 8.5, roughly 32 times more energy than the other neotectonic settings. The large anomalous magnitudes of energy release in convergent margins are attributed to the transfer of inter-plate stress to the upper-plate, where convergent elastic strain is accumulated during interseismic intervals. The large volumes of rock that accumulate the elastic strain in the upper-plates of convergent zones are defined here as primary seismogenic structures. Several datasets of 1) modern upper-plate convergent strain, 2) historical earthquakes, 3) modern upper-plate vertical displacements, and 4) recent inter-plate events of Episodic Tremor and Slip (ETS) are compared to establish the extent of the primary seismogenic structure in the Cascadia convergent zone. The across-margin extents of 1) significant convergent strain, 2) margin-parallel bands of vertical displacement, 3) historical seismicity and 4) ETS events, representing inter-plate coupling and shear stress transfer to strain accumulation in the upper-plate, are used to map the width of the primary seismogenic structure. The across-margin width of the primary seismogenic structure in the central Cascadia margin ranges from 300 km in the south-central margin to 450 km in the north-central margin, as mapped landward from the buried trench. A broad source region of coseismic energy release in the Cascadia primary seismogenic structure (300 - 450 km width) could yield stronger shaking in interior metropolitan centers from a future major rupture of the mega-thrust than has been modeled from a narrow “locked” zone located offshore under the outer 展开更多

This paper argues that historians have all but ignored the study of rivers and their impact on the development of human society. Apart from a somewhat terse acknowledgment of the importance of rivers in the development of ancient civilizations, from the Huang He to the Ganges, the Nile, and the Amazon, historians have by and large limited themselves to studying individual rivers, while ignoring the potential of comparative analysis of rivers. I call for a broader engagement by historians of all aspects of rivers, including their role in transportation, fishing, agriculture, industry, recreation, and the environment, peopled cultural response to rivers, and the legal regimes that have grown up around them, with special reference to the role of rivers as political boundaries. 展开更多

A 57-year-old woman underwent abdominal surgery with a subarachnoid block supplemented by “light” general endotracheal anesthesia consisting of a propofol infusion and a sub-MAC concentration of sevoflurane. The previous case in the same operating room had involved a malignant hyperthermia-susceptible patient, and charcoal filters had been placed in the breathing circuit as a precautionary measure. Because it had not been used on the evening beforehand, the circuit with filters was left in situ with a strip of tape indicating that it was clean. The woman’s anesthesiologist assumed that these filters were heat and moisture exchanger filters in an unused circuit and therefore did not remove them. Subsequently, the patient had awareness with intraoperative recall. This case highlights the potential for inadvertent use of activated charcoal filters with potentially catastrophic results. Such unintended utilization of these products likely can be minimized by improved labeling techniques. 展开更多

Effect of N2 and O2 on OH radical production in an atmospheric helium microwave plasma jet

UV-pulsed laser cavity ringdown spectroscopy of the hydroxyl radical OH(A–X)(0–0)band in the wavelength range of 306–310 nm was employed to determine absolute number densities of OH in the atmospheric helium plasma jets generated by a 2.45 GHz microwave plasma source.The effect of the addition of molecular gases N2 and O2 to He plasma jets on OH generation was studied.Optical emission spectroscopy was simultaneously employed to monitor reactive plasma species.Stark broadening of the hydrogen Balmer emission line(Hβ)was used to estimate the electron density ne in the jets.For both He/N2 and He/O2 jets,ne was estimated to be on the order of 10^15 cm^−3.The effects of plasma power and gas flow rate were also studied.With increase in N2 and O2 flow rates,ne tended to decrease.Gas temperature in the He/O2 plasma jets was elevated compared to the temperatures in the pure He and He/N2 plasma jets.The highest OH densities in the He/N2 and He/O2 plasma jets were determined to be 1.0×10^16 molecules/cm^3 at x=4 mm(from the jet orifice)and 1.8×10^16 molecules/cm^3 at x=3 mm,respectively.Electron impact dissociation of water and water ion dissociative recombination were the dominant reaction pathways,respectively,for OH formation within the jet column and in the downstream and far downstream regions.The presence of strong emissions of the N2^+ bands in both He/N2 and He/O2 plasma jets,as against the absence of the N2^+ emissions in the Ar plasma jets,suggests that the Penning ionization process is a key reaction channel leading to the formation of N2^+ in these He plasma jets. 展开更多

BACKGROUND A variety of immune-modulating drugs are becoming increasingly used for various cancers.Despite increasing indications and improved efficacy,they are often associated with a wide variety of immune mediated adverse events including colitis that may be refractory to conventional therapy.Although these drugs are being more commonly used by Hematologists and Oncologists,there are still many gastroenterologists who are not familiar with the incidence and natural history of gastrointestinal immune-mediated side effects,as well as the role of infliximab in the management of this condition.CASE SUMMARY We report a case of a 63-year-old male with a history of metastatic renal cell carcinoma who presented to our hospital with severe diarrhea.The patient had received his third combination infusion of the anti-CTLA-4 monoclonal antibody Ipilimumab and the immune checkpoint inhibitor Nivolumab and developed severe watery non-bloody diarrhea the same day.He presented to the hospital where he was found to be severely dehydrated and in acute renal failure.An extensive workup was negative for infectious etiologies and he was initiated on high dose intravenous steroids.However,he continued to worsen.A colonoscopy was performed and revealed no endoscopic evidence of inflammation.Random biopsies for histology were obtained which showed mild colitis,and were negative for Cytomegalovirus and Herpes Simplex Virus.He was diagnosed with severe steroid-refractory colitis induced by Ipilimumab and Nivolumab and was initiated on Infliximab.He responded promptly to it and his diarrhea resolved the next day with progressive resolution of his renal impairment.On follow up his gastrointestinal side symptoms did not recur.CONCLUSION Given the increasing use of immune therapy in a variety of cancers,it is important for gastroenterologists to be familiar with their gastrointestinal side effects and comfortable with their management,including prescribing infliximab. 展开更多

Biomass energy has become an important measure to alleviate ecological environment security and energy supply security in China.Emergy accounting method is used to analyze and evaluate economy,environment,and sustainability of corn straw generation system,which includes corn planting subsystem,collection and transportation subsystem,and corn straw power generation subsystem.The key substances that need to be optimized in the system are identified by using sensitivity analysis.Based on the position of key substances in the system,emergy accounting optimization methodology is conducted.Corresponding optimization design scheme is proposed based on the '3R' (reduce,reuse,and recycle) principle of circular economy.Current study shows that emergy yield ratio,emergy investment ratio,environmental loading ratio,and emergy sustainability index of the corn straw power generation system are 3.69,2.68,1.61,and 2.29,respectively,which are better than wind power generation system and thermal power generation system.In addition,it is proposed that the fertilizer alternative plan and the transportation redesign plan can replace 50,000 tons potash and reduce equipment resources by 20% and energy resources input by 30%.Economy and sustainable development of the system is significantly improved.The optimization design method based on the '3R' principle proposed in this paper can provide a useful reference for the research in the field of resource recycling. 展开更多

Winter Rain versus Snow in Headwater Catchments: Responses of an Unconfined Pumice Aquifer, South-Central Oregon, USA 预览

Winter precipitation in two headwaters catchments (elevation ~1600 m) in the rain shadow of the Cascades volcanic arc in south-central Oregon normally falls as snow. However, in water year 2015, winter precipitation fell mainly as rain. An eight year study of the unconfined pumice aquifer allowed inter-annual comparison of groundwater recharge during the freshet and discharge during the growing season. During these water years precipitation ranged from 67% (WY2014) to 132% (WY2017) of the 30 year average, and included the rain dominated winter of WY2015 when precipitation during the water year was 98% of the 30 year average. Change in storage in the pumice aquifer was estimated from thickness of the pumice deposit and depth to water table from the ground surface. Measurements were made where 1) the pumice aquifer was exposed at the surface;2) where the aquifer was partially eroded and overlain by either alluvium or lacustrine glassy silt to fine sand;3) fens where the partially eroded aquifer was overlain by peat;and 4) monitoring wells drilled through the pumice aquifer into bedrock. In all settings, groundwater storage in the pumice aquifer following the rain-dominated winter of WY2015 was similar or less than storage following the drought of WY2014 when winter precipitation fell as snow. Storage at the end of WY2014 and WY2015 was the least observed in the eight year study. Winter-time rain during WY2015 produced runoff rather than storage in snow pack. Runoff conveyed from the catchments by flow in stream reaches normally dry from late summer through the winter months. Rain-dominated winter precipitation stresses the perched pumice aquifer. Winter storms starting as rain and turning late to snow and ground-freezing temperatures lead to runoff during the next rain-dominated precipitation event. These patterns produced stream flow in channels that are commonly dry during the winter, reduced near-surface groundwater storage in the pumice aquifer, muted springtime freshet, and stressing of groundwater-dependent e 展开更多

Cervical dystonia (CD) is a condition that typically presents with cervical muscle spasm, producing head tilt and cervical rotation. CD is most often idiopathic, however, in a small number of patients, CD occurs within one day to one year after mild to severe trauma. This type of CD is further classified as posttraumatic CD. OnabotulinumtoxinA (Botox) injections are considered to be a controversial treatment for posttraumatic CD and have produced variable result. This report describes the case of a 32-year-old female presenting with a two year history of posttraumatic CD and associated head, neck, and shoulder pain after obtaining a severe head injury during a motorcycle accident. OnabotulinumtoxinA was used to successfully treat her posttraumatic CD muscle spasms and associated chronic pain. Three months after her first and second ONA treatments, the patient reported at least 50% improvement in her overall pain symptoms and a noticeable reduction in cervical paraspinal muscle spasms. 展开更多

Performance and Optimization of a Small Hybrid Solar-Thermal Collector 预览

A hybrid solar collector was designed to investigate the effects of combining two different solar collector techniques on the overall collector’s effectiveness. While most solar collectors focus only on one solar collection method, the small hybrid system uses a flat plate collector in conjunction with five evacuated tubes to absorb the most energy possible from both direct and diffuse solar radiation. Data was collected over four months while the system operated at different flow rates and with various levels of available insolation from the sun to evaluate the performance of the solar collector. To understand the relative contribution of the flat plate collector and the evacuated tubes, temperature differences across each part of the system were measured. The results indicate the average first law efficiency of the hybrid system is 43.3%, significantly higher than the performance of the flat plate alone. An exergy analysis was performed for this system to assess the performance of the flat plate system by itself. Results of the second law analysis were comparable to the exergetic efficiencies of other experimental collectors, around 4%. Though the efficiencies were in the expected range, they reveal that further improvements to the system are possible. 展开更多

Modern horizontal strain (2006-2016) measured along 56 new and 108 previously published GPS station baselines are used to establish the length (800 km) and width (300 - 400 km) of the central Cascadia convergent margin seismogenic structure. Across-margin (west-east) annual rates of shortening range from 10﹣9 a﹣1 at the eastern (landward) limit of the central Cascadia seismogenic structure to 10﹣7 a﹣1 along the western onshore portion of the interplate zone. Relatively high shortening strain rates (10﹣8 a﹣1 to 10﹣7 a﹣1) are also measured in western transects from the northern (Explorer plate) and southern (Gorda plate) segments of the convergent margin, demonstrating that the full length of the margin (1300 km length) is currently capable of sustaining and/or initiating a major great earthquake. Vertical GPS velocities are averaged over the last decade at 321 stations to map patterns of uplift (0 - 5 mm yr﹣1) and subsidence (0 - 9 mm yr﹣1) relative to the study area mean. Along-margin belts of relative uplift and subsidence, respectively, are approximately associated with Coast Ranges and the Cascade volcanic arc. However, the vertical velocity data are locally heterogeneous, demonstrating patchy “anomalies” within the larger along-margin belts. A large coastal subsidence anomaly occurs in southwest Washington where the modern short-term trend is reversed from the long-term (~200 yr) tidal marsh record of coastal uplift since the last co-seismic subsidence event (AD1700). The modern vertical displacements represent a late stage of the current inter-seismic interval. If the horizontal strain is considered largely or fully elastic, extrapolating the modern strain rates over the last 100 years show the accumulated strains would be similar in magnitude to the observed co-seismic strains resulting from the Tōhoku, Japan, Mw 9.0 earthquake in 2011.
