薏苡组培快繁技术及遗传变异研究 认领

Study on Tissue Culture and Rapid Propagation Technique and Genetic Variation of Coix Lacryma-jobi

摘要 薏苡(Coix Lacryma-jobi L.)为禾本科药食兼用作物,具有较高的营养及药用价值。本研究以贵州省地方薏苡品种兴仁小白壳为材料,以芽顶端分生组织为外植体,探究外植体最佳消毒条件、最佳丛生芽增殖培养基、生根培养基配方以及适合幼苗移栽成活的基质配比,并采用SSR分子标记技术对继代兴仁小白壳组培苗进行遗传变异分析。结果表明:兴仁小白壳茎尖最佳消毒条件为75%酒精消毒30 s、0.2%升汞处理7 min,最佳增殖培养基为MS+0.5 mg·L^-1 NAA+9 mg·L^-16-BA,增值系数为2.9,最佳生根培养基为1/2 MS+0.5 mg·L^-1 NAA,生根率为100%,移栽条件为V营养土∶V珍珠岩=4∶1时成活率达95%,继代培养6次后的植株与亲本的遗传特性均保持一致。本研究建立的兴仁小白壳组培快繁体系为优质薏苡种质资源的保存提供了技术参考。 Coix Lacryma-jobi is a kind of medicine and food crop with high nutritional and medicinal value.In this study,the meristems at the top of buds of Guizhou local C.Lacryma-jobi variety Xinrenxiaobaike were used as explants to explore the optimal disinfection conditions of the explants,the optimal culture medium for the proliferation of fascicled buds,rooting medium and the matrix ratio suitable for the survival of the seedlings.Results showed that the best disinfection condition for Xinrenxiaobaike was disinfection with 75%alcohol for 30 s,followed by treatment with 0.2% mercury bichloride for 7 min.And the best proliferation medium was MS+0.5 mg·L^-1 NAA+9 mg·L^-16-BA,with growth coefficient being 2.9,while the best rooting medium was 1/2 MS+0.5 mg·L^-1 NAA,with rooting rate bing 100%.The best transplanting matrix was 4 nutrient soil∶1 perlite(V∶V)with 95%survival rate,and the genetic characteristics of the plants after 6 times of subculture were consistent with that of their parents.Tissue culture and rapid propagation system established in this paper provided a technical reference for the preservation of quality C.Lacryma-jobi germplasm resources.
